胎盘组织miR-496表达水平与子痫前期关联性研究 被引量:1

摘要 目的探讨胎盘组织miR-496表达水平与子痫前期相关性。方法选取2015年5月-2017年7月间江苏省苏北人民医院和扬州大学临床医学院收治的120例孕产妇作为研究对象,分为正常孕产妇30例(A组),妊娠期高血压30例(B组),子痫前期轻度30例(C组),子痫前期重度30例(D组) 4组;使用HE染色对各组胎盘组织进行检测,并采用RT-PCR检测胎盘组织中miR-496及GAPDH表达情况。结果胎盘组织可见各级绒毛干不同程度动脉粥样硬化,并且多例合并合胞体结节增多;单例可见合胞体滋养层空泡变性;轻度子痫、重度子痫、妊娠高血压及正常妊娠孕产妇miR-496/GAPDH水平比较差异有统计学意义(P<0. 05),且重度子痫组显著高于轻度子痫组、妊娠高血压组及正常妊娠组(P<0. 05),轻度子痫组显著高于正常妊娠组(P<0. 05);胎盘组织中miR-496表达水平与子痫前期病情正性相关,等级相关系数r=0. 738 (P<0. 05)。结论随着子痫前期患者病情加重其胎盘组织中miR-496表达水平也随之升高,可有效反应患者病情。 Objective To investigate the correlation of miR-496 expression in placental tissue with preeclampsia. Methods From May 2015 to July 2017, 120 cases in Subei People's Hospital of Jiangsu Province and Clinical Medicine School of Yangzhou University were selected as the study objects, including 30 cases of normal pregnant women (group A) , 30 cases of hypertensive disorder complicating preg- nancy ( group B) , 30 cases of mild preeclampsia ( group C) , 30 cases of severe preeclampsia ( group D) ; the tissue staining was detected u- sing HE, and the expressions of miR-496 and GAPDH in placenta were detected by RT-PCR. Results Placenta stem villi could be seen at all levels of different degrees of atheroselerosis, and many patients with syncytial nodules increased; single cases showed syntrophoblast de- generation; miR-496/GAPDH levels showed significant difference in mild preeclampsia and severe preeclampsia, hypertensive disorder com- plicating pregnancy and normal pregnant women (P〈0. 05 ) , and severe preeclampsia group was significantly higher than that of mild pre- eclampsia group, gestational hypertension group and normal pregnancy group (P〈0. 05 ) , mild preeclampsia group was significantly higher than that of normal pregnancy group ( P〈0. 05 ) ; the level of miR-496 expression in placenta tissue was associated with positive preeclamp- sia, grade correlation coefficient r=O. 738 (P〈0. 05) . Conclusion With the aggravation of the preeclampsia, the level of miR-496 ex- pression in placenta tissue is also increased, which can effectively respond to the patient's condition.
