Effect of interleukin-4 on modulating osteoclast differentiation and function in mouse bone marrow culture

摘要 目的 :观察IL 4对诱导骨髓干细胞分化形成破骨细胞的作用。方法 :用终浓度为 10 -8mol·L-1的 1,2 5(OH) 2 D3 诱导成年小鼠骨髓干细胞分化形成破骨细胞 ,在此过程中加入不同质量浓度梯度的IL 4(0、0 .1、1、10 μg·L-1)以及用IL 4的抗体中和IL 4。取IL 4(1μg·L-1)处理过的第 6天的培养上清液 ,先用抗IL 4抗体中和残余IL 4,再以体积分数 1% ,10 % ,2 0 % ,0加入另一新培养 4d含有骨片的骨髓细胞培养体系中 ,计数骨片上骨吸收陷窝数及面积 ,玻片上抗酒石酸酸性磷酸酶 (tartrate resistantacidphosphatase ,TRAP)阳性多核细胞和单核细胞数。结果 :随IL 4(质量 )浓度的增加 ,骨片上形成的骨吸收陷窝及面积数 ,玻片上的TRAP阳性细胞数呈量的依赖性的减少 ,加抗体组抵消了IL 4的抑制作用 ,加条件培养液组和未加组上述参数没有显著差别。结论 :IL 4具有抑制骨髓干细胞分化形成破骨细胞的作用 ,这种作用不是通过使其它细胞形成可溶性因子 ,可能是直接作用于靶细胞 (能分化为破骨细胞和巨噬细胞的前体细胞 ) ,使定向分化为TRAP阳性单核细胞的前体细胞转向分化为巨噬细胞 ,破骨细胞数目减少 。 Objective: To observe the effect of IL 4 on murine hematopoietic stem cells differentiating into osteoclasts in mouse bone marrow induced by 1,25(OH) 2D 3. Methods: In the process of osteoclast formation from mouse bone marrow induced by 1,25(OH) 2D 3(10 -8 mol·L -1 ), IL 4 withdifferent concentration (0 μg·L -1 , 0.1 μg·L -1 , 1 μg·L -1 , 10 μg·L -1 ) was added and anothergroup was designed with anti IL 4 antidodies. The cell free supernants of cultures treated with IL 4(1μg·L -1 ) were preincubated with anti IL 4 antibodies (1 g·L -1 )to block any residual active IL 4 andadded to other fresh bone marrow cultures on day 4(0% 20% vol/vol). The number of bone resorption pits and area on bone slices and the number of tartrate resistant acid phosphatase (TRAP) positive cells on glass slices were counted. Results: The addition of IL 4(0.1 10 μg·L -1 )at the startof the culture period induced a dose dependent decrease in the number of bone resorption pits, area; and TRAP positive cells. In the anti IL 4 neutralizing group and conditioned medium groups, theparameters which were the same as the above, compared with the control group, had no significantdifference in statistics. Conclusion: IL 4 could inhibit osteoclast formation from hepatopoietic stemcells in vitro . Such an effect did not seem to be mediated by release of other soluble factors from othercells, and it might directly acts on uncommitted precursors able to differentiate toward both osteoclastsand macrophages. The cytokine induced a preferential differention toward the macrophage phenotype, thus decreasing the generation of osteoclasts and inhibiting bone resorption.
