摘要
目的 了解 TIMP- 1和 TIMP- 2在肝硬化患者肝组织中的表达状态 ,探讨 TIMP- 1和 TIMP- 2在肝硬化发病机制中的作用 .方法 以抗 TIMP- 1和 TIMP- 2单克隆抗体(Mc Ab)为试剂 ,采用免疫组织化学法检测肝硬化患者肝组织中 TIMP- 1,TIMP- 2相关抗原的表达 .结果 30例肝硬化患者肝组织中 TIMP- 1和 TIMP- 2相关抗原表达的阳性率为10 0 % ,,TIMP- 1阳性的肝组织中 TIMP- 2亦为阳性 ,且TIMP- 1强度高于 TIMP- 2 ,而正常肝组织无 1例表达阳性 .TIMP- 1和 TIMP- 2抗原表达阳性信号主要位于肝细胞胞质中 ,未见细胞核表达 .结论 肝硬化患者肝组织中的肝细胞中存在 TIMP- 1和 TIMP- 2相关抗原表达 ,TIMP- 1和 TIMP-2与肝病的进展相关 ,它通过抑制 MMPs的活性 ,使得 ECM降解减少而导致肝纤维化 ,以至肝硬化 .
AIM To investigate the distribution of tissue inhibitors of metallproteinanses 1 and 2 (TIMP 1 and TIMP 2) liver cirrhosis tissue of patients and discuss the function of TIMP 1 and TIMP 2 in pathogenesis of liver cirrhosis. METHODS TIMP 1 or TIMP 2 related antigens were detected in paraffin embeded liver tissue of biopsy patients with liver cirrhosis by immunohistochemical method using specific monoclonal antibody (McAb). RESULTS Among 30 samples, 100% had been detected out TIMP 1 and TIMP 2 related antigen in the liver cirrhosis. Both expressions of TIMP 1 and TIMP 2 were positive, and expression of TIMP 1 was higher than TIMP 2. None of normal liver tissue was positive. The staining signal of TIMP 1 and TIMP 2 related antigen was mainly located in cytoplasm of liver cells. There was no signal in nuclear of liver cells. CONCLUSION These studies suggest that both TIMP 1 and TIMP 2 related antigens may exist in liver cirrhosis tissue of patients. They are correlated with development. They reduce excellular matrix (ECM) and lead to liver fibrosis and liver cirrhosis by restraining the activity of matrix metalloproteinase.
