Expression and relationship of Beclinl and Bcl2 in invasive pituitary adenomas

摘要 目的检测Beclin1和Bcl2在侵袭性垂体腺瘤组织中的表达,探讨其与垂体腺瘤侵袭性的关系及2者之间的相关性。方法61例垂体腺瘤手术标本按侵袭性垂体腺瘤综合判定方法分为侵袭组(32例)和非侵袭组(29例),采用免疫荧光检测Beclin1和Bcl2蛋白的表达,RT-PCR法检测Beclin1和Bcl2mRNA的表达,分析侵袭组和非侵袭组Beclin1和Bcl2表达水平的差异和相关性。结果侵袭组Beclin1蛋白和mRNA表达均低于非侵袭组(P〈0.01);而Bcl2蛋白和mRNA表达均高于非侵袭组(P〈0.01)。Pearson相关分析显示:侵袭组中Beclin1和和Bcl2mRNA的表达呈负相关(r=-0.42,P=0.028)。结论Beclin1在侵袭性垂体腺瘤中的表达下降,垂体腺瘤的侵袭性与Bcl2蛋白及mRNA的高表达,Beclin1蛋白和mRNA的低表达密切相关;自噬活性的减低可能导致垂体腺瘤侵袭性增强,而其活性减低可能是Beclin1和Bcl2相互作用的结果。 Objective To detect the expression of Beclinl and Bcl2 in invasive pituitary adenomas and to explore the relationship of Beclin1 and Bel2 in invasive pituitary adenomas and the relativity between the 2 genes. Methods 61 specimens were classified into invasive group (32 cases) and non-invasive group (29 cases) according to the comprehensive evaluation of invasive pituitary adenomas. Immnnofluorescence analysis and RT-PCR were adopted respectively to detect the protein and mRNA expressions of Beclinl and Bel2. The difference and relativity of Beclinl and Bcl2 expression in invasive group and non-invasive group were analyzed. Results 32 specimens of pituitary adenoma were invasive and 29 were non-invasive. Beclin1 protein and mRNA expressions were lower in the invasive group than in the non-invasive group ( P 〈 0. 01 ). Bcl2 protein and mRNA expressions were higher in the invasive group than in the non-invasive group ( P 〈 0. 01 ). Pearson related analysis showed that Beclinl mRNA expression was negtively correlated with Bcl2 mRNA expression in the invasive group ( r = - 0. 42, P = 0. 028). Conclusions Beclinl expression is decreased in invasive pituitary adenomas. The invasiveness of pituitary adenoma is closely related to the high expression of Bcl2 protein and mRNA, and the low expression of Beclinl protein and mRNA. The inhibition of the autophagy may lead to the enhancement of the invasiveness of pituitary adenomas and that inhibition may come from the interaction of Beelinl and Bcl2.
