摘要
利用MTT法检测番木瓜种子中的异硫氰酸苄酯(BITC)在体外对肝癌细胞等6种常见癌细胞的生长抑制率。结果表明,番木瓜种子提取物中BITC的浓度达到5μmol.L-1(相当于0.745mg.L-1)时,对人肝癌HepG2细胞、人肺癌A549细胞、人乳腺癌MCF-7细胞、人大肠癌HCT-8细胞、人宫颈癌HeLa细胞、人前列腺癌DU-145细胞的生长抑制率均达到70%以上,其中,人肝癌细胞HepG2最敏感,cBITC=2.5μmol.L-1时抑制率可达87%,在cBITC=10μmol.L-1和5.0μmol.L-1时,人大肠癌HCT-8细胞的生长抑制率几乎相同。因此,BITC具有良好的抑癌效果。
Benzyl isothiocyanate (BITC) was extracted from papaya seeds (Carica papaya L. ) and used to determine its inhibitory effect on the cell growth of 6 common cancers in vitro including liver cancer by using the MTr method. The results showed the papaya seed extract with BITC at the concentration of up to 5 μmol · L-1 ( equivalent to 0.745 mg · L-1 ) inhibited more than 70% of the cell growth of a human liver cancer cell line HepG2, a lung cancer cell line A549, a breast cancer cell line MCF-7, a colon cancer cell line HCT-8, a cervical cancer cell line Hela and a prostate cancer cell line DU-145. Of them the HepG2 liver cancer cells were the most sensitive to BITC which gave an 87% inhibition over the ceils at the concentration 2.5 μmol · L-1. HCT-8 colon cancer cells had a similar inhibition rate when treated with BITC at both 5.0 and 10μmol· L-1. This showed that higher concentration of BITC in the extract of papaya seeds was similar to the lower concentration (5.0 μmol · L-1 ) in inhibition of the growth of the cancer cells.
