摘要
目的:通过观察养精种玉汤对过多雄激素诱导卵泡胰岛素抵抗的影响,初步探讨其促卵泡发育的机制。方法:体外培养猪卵泡,经过多雄激素作用48 h后,检测猪卵泡对葡萄糖的摄取量和卵泡培养液上清残糖量。分别检测正常组、抵抗组和复方组卵泡胰岛素受体底物-1(IRS-1)和葡萄糖转运体4(GLUT4)蛋白表达水平以及卵泡培养液上清残糖量。结果:过多雄激素作用48 h后,卵泡对葡萄糖的摄取量较正常组下降39.56%(P<0.01),卵泡培养液上清残糖较正常组升高29.08%(P<0.01)。当卵泡胰岛素抵抗时,IRS-1与GLUT4蛋白表达量较正常组分别降低74%与94.56%(P<0.01);加入养精种玉汤复方改善胰岛素抵抗后,复方组IRS-1与GLUT4蛋白表达量较抵抗组分别升高2倍与5倍(P<0.01),卵泡培养液上清残糖较抵抗组降低33.6%(P<0.001)。结论:过多雄激素能诱发卵泡胰岛素抵抗,养精种玉汤能改善卵泡胰岛素抵抗,其机制与IRS-1与GLUT4表达量有关。
Objective: To discusses the development of the follicle stimulating mechanism by observing Yangjing Zhongyu decoction reducing insulin resistance induced by excess androgen in follicle. Method: (1 Follicles from porcine ovary were isolated and cultured in vitro. The follicles were induced by excess androgen for 48 h. Then the follicle glucose intake and glucose content were measured in culture solution. (2) The protein expression of insulin receptor substrate-1 ( IRS-1 ) and glucose transporter 4 (GLUT4) and glucose content were detect in culture solution of each group. Result: After being treated by excess androgen for 48 h, follicle glucose intake was decreased about 39. 56% (P 〈0.01 ), the glucose content in culture solution was increased for about 29.08% (P 〈0.01 ). The levels of IRS-1 and GLUT4 protein expression was decreased about 74% and 94. 56% in follicles induced by excess androgen (P 〈 0.01, P 〈 0.01). However, the protein expression of IRS-1 and GLUT4 was increased 2 times and 5 times (P 〈0.01 ), the glucose content in culture solution was decreased for about 33.6% (P 〈 0. 001 ) after treated with Yangjing Zhongyu decoction. Conclusion: The mechanism of excess androgen can induce in sulin resistance and Yangjing Zhongyu decoction reduce insulin resistance in follicle which may be related to the levels of IRS-1 and GLUT4 nrotein exnression.
