摘要
目的观察吴茱萸次碱对重症急性胰腺炎(SAP)的干预作用以及与乌司他丁联用的疗效。方法 50只SD大鼠随机均分为5组:假手术组,模型组,乌司他丁组,吴茱萸次碱组,乌司他丁+吴茱萸次碱组,每组10只。除假手术组外,其余各组大鼠以5%牛磺胆酸钠胰胆管逆行注射制作SAP模型并给予相应的处理。造模后24 h,对大鼠胰腺组织进行病理学检查,并检测血清淀粉酶活性和TNF-α和IL-6水平。结果除假手术外,各用药组病理学评分均明显低于模型组,且联合用药组评分最低(均P<0.05)。各用药组血清淀粉酶活性显著低于模型组(P<0.05),且联合用药组评分最低(均P<0.05)。各实验组和模型组大鼠血清TNF-α和IL-6水平均明显高于假手术组(均P<0.05),且联合用药组评分最高(均P<0.05)。结论吴茱萸次碱对大鼠SAP具有治疗作用,其机制可能与改善胰腺组织微循环有关;吴茱萸次碱与乌司他丁联用能明显增强对SAP的疗效。
【Objective】 To investigate the interventional effect of rutecarpine on rats with severe acute pancreatitis(SAP) and the combined efficacy of rutecarpine and ulinastatin.【Methods】 Fifty SD rats were equally randomized into sham operation group,model group,ulinastatin group,rutecarpine group and ulinastatin plus rutecarpine group.Each group had ten rats.Except for the sham group,all rats underwent retrograde cholangiopancreatic duct injection of 5% sodium taurocholate to induce SAP model,and then were given their regimens.Twenty-four hours after the model establishment,rats were sacrificed,and the samples of blood and pancreatic tissue were harvested.The pathological changes of the pancreatic tissues were examined,the serum levels of amylase was detected by using automatic biochemistry analyzer,level of TNF-α and IL-6 were measured by Elisa kits.【Results】 Except for the sham group,the pathological scores of all the treatment groups were significantly lower than that of the model group,and the combined treatment group showed the lowest score(all P 〈0.05).The serum amylase level of all the treatment groups were significantly lower than that of the model group,and the combined treatment group showed the lowest serum amylase level(both P 〈0.05).The serum levels of TNF-α and IL-6 of all the treatment groups were significantly higher than that of the model group(all P 〈0.05),and the combined treatment group showed the highest serum levels of TNF-α and IL-6(both P 〈0.05).【Conclusion】 Rutecarpine has therapeutic effect on SAP of rats,and the mechanism maybe associated with its improving the microcirculation of the pancreatic tissue.The combination treatment of ulinastatin and rutecarpine has enhanced efficacy in treatment of SAP.
