摘要
目的检测多发性骨髓瘤(MM)患者外周血CD4+IL-17A+辅助性T细胞(Th17细胞)/CD4+CD2+5Foxp3+调节性T细胞(Treg细胞)比率及相关细胞因子,分析其相关性。方法采用流式细胞术检测MM组初诊患者外周血Th17、Treg细胞占CD4+T细胞比例,ELISA法检测MM组初诊患者血清IL-17、IL-6、IL-23、TGF-β、IL-10水平。结果 MM患者外周血Th17细胞比率升高,Treg细胞比率降低,血清IL-6、IL-17与IL-23水平明显升高;MM患者外周血Th17细胞与Treg细胞含量呈负相关,Th17细胞含量与血清IL-6、IL-17、IL-23水平均呈正相关,Treg细胞含量与血清中IL-6、IL-17、IL-23水平均呈负相关。结论 MM患者存在Th17/Treg细胞失衡,IL-6、IL-17、IL-23影响两者表达,在MM发生发展中起重要作用。
Objective To observe the proportions of CD+4IL-17A+helper T cells(Th17 cells)and CD+4CD+25Foxp3+regulatory T cells(Tregs)and the related cytokines in the peripheral blood of multiple myeloma patients and to analyze their relationship.Methods The proportions of Treg and Th17cells in peripheral CD+4T cells were determined using flow cytometry.The expressions of peripheral serum IL-17,IL-6,IL-23,TGF-β,and IL-10 in untreated multiple myeloma patients with untreated were determined with enzyme-linked immunosorbent assay(ELISA).Results The proportion of Th17 cells increased,the proportion of Treg cells decreased,and the serum levels of IL-17,IL-6,and IL-23 increased in patients with untreated multiple myeloma.Th17 cells were negative correlated with Treg cells,whereas Th17 cells were positively correlated with IL-17,IL-6,and IL-23 and Treg cells were negatively correlated with these cytokines.Conclusion Imbalance between Treg and Th17 cells exist in multiple myeloma patients.IL-17,IL-6,and IL-23 can effect the expressions of Treg and Th17 cells and thus play roles in the development of multiple myeloma.
