九香止泻片对腹腔注射致死量细菌小鼠IL-8影响的实验研究

Experimental study on effect of Jiuxiang Zhixie tablets on interlukin-8 level in mice intraperitoneally injected with lethal doses of bacteria

摘要 目的:观察九香止泻片对腹腔注射致死量大肠杆菌、痢疾杆菌小鼠白介素-8(IL-8)的影响,探讨其治疗急性感染性腹泻的作用机制。方法:分别给小鼠腹腔注射致死量大肠杆菌、痢疾杆菌等不同细菌菌悬液制作腹泻感染动物模型,除模型组小鼠在造模成功后采用无菌操作心脏采血外,其余各组在处死动物之前采用无菌操作心脏采血,然后采用双抗体夹心酶联免疫吸附(ELISA)法检测IL-8。结果:与模型组比较,九香止泻片各剂量组具有明显降低感染模型小鼠炎症因子IL-8含量的作用(均P﹤0.05),且九香止泻片高剂量组与肠康片组比较,差异具有统计学意义(均P﹤0.05)。结论:提示九香止泻片可能参与炎性介质的调节,调控细胞免疫功能可能是其治疗急性感染性腹泻的机制之一。 Objective : To observe the effect of Jiuxiang Zhixie tablets on the interlukin - 8 ( IL - 8 ) level in the mice intraperitone- ally injected with lethal doses of Eseherichia coli and Shigella, and to investigate the mechanism of Jiuxiang Zhixie tablets in treatment of acute infectious diarrhea. Methods : Suspensions of Escherichia coli and Shigella were intraperitoneally injected into mice at lethal doses to establish an animal model of acute infectious diarrhea. After the model was established, blood was sampled aseptically from the heart of the mice in a model group. In treatment groups, which received treatment with different doses of Jiuxiang Zhixie tablets or Changkang tablets, blood was sampled aseptically from the heart of mice before the mice were killed, and then double - antibody sand- wich enzyme - linked immunosorbent assay was used to measure the IL - 8 level. Results : Compared with the model group, the Jiux- iang Zhixie tablet treatment groups showed significantly decreased IL - 8 level ( P 〈 0.05 ). A significant difference was also found be- tween the high - dose Jiuxiang Zhixie tablet treatment group and Changkang tablet treatment group ( P 〈 0.05 ). Conclusion : Jiuxiang Zhixie tablets may play a role in regulation of inflammatory mediators, and it might treat acute infectious diarrhea through regulating cellular immune function.
