骨康含药血清对细胞共育体系中OPG、RANKL的影响 被引量:10

Effects of Serum Containing Gukang on Osteoprotegerin and Receptor Activator of Nuclear Factor-κB Ligand in the Cell Co-culture System

摘要 目的:研究骨康含药血清对细胞共育体系中OPG、RANKL的影响。方法:以成骨-破骨细胞共培养液上清中OPG、RANKL及其比值为指标进行实验观察。结果:加入血清培养后,自第3天起骨康组OPG含量明显高于同时期空白组(P<0.01),而空白组培养过程中无明显变化。骨康组RANKL含量第7天明显下降(P<0.05),而空白组无明显变化(P>0.05)。自第3天起骨康组OPG/RANKL比值显著高于空白组(P<0.05,P<0.01)。在培养第5、7天骨康组OPG/RANKL比值较初始比值明显增大(P<0.05),而空白组在培养过程中无明显变化。结论:骨康含药血清对成骨细胞和破骨细胞均有影响,可改变细胞生存环境中OPG/RANKL的比例,间接影响破骨细胞的功能活性。 Objective:To study the effect of serum containing Gukang on osteoprotegerin(OPG)and Receptor Activator of Nuclear Factor-κB Ligand(RANKL)in the cell co-culture system.Methods:We observed changes of OPG,RANKL and their ratio in osteoblast-osteoclast culture supernatant.Results:After adding serum-free medium,since the 3rd day,OPG content in Gukang group was significantly higher than that in the blank group(P〈0.01),RANKL content in Gukang group was significantly lower than that in the blank group on the 7th day(P〈0.05),but OPG and RANKL in the blank group had no significant change in the culture period.Since the 3rd day,the ratio of OPG/RANKL in Gukang group was significantly higher than that in the blank group(P〈0.05,P〈0.01),the ratio of OPG/RANKL on the 5th and 7th day was higher than that on the 1st day in Gukang group(P〈0.05),but the ratio of OPG/RANKL in the blank group had no significant change in the culture period.Conclusion:Serum containing Gukang is effective on osteoblast and osteoclast,it can regulate the ratio of OPG/RANKL in cell survival environment,and indirectly affect the functional activity of osteoclast.
