摘要
目的通过观察骨康含药血清对成骨细胞IL-1、IL-6和TNF-α表达的影响,探讨其治疗骨质疏松的作用机制。方法体外分离、培养成骨细胞,实验分为3组:正常血清组、中药骨康血清组和雌二醇含药血清组。采用酶联免疫吸附法,检测成骨细胞IL-1、IL-6和TNF-α表达。结果中药骨康含药血清组IL-1、IL-6和TNF-α含量显著低于空白对照组(P<0.05),中药骨康含药血清组与雌二醇含药血清组IL-1、IL-6和TNF-α的含量无明显差异。结论在去势状态下,中药骨康可能是抑制IL-1、IL-6和TNF-α分泌,而达到治疗骨质疏松症的作用。
Objective To investigate the effects of serum containing substances of a Chinese traditional drug, Gukan, on expression of IL-1, IL-6 and TNF-alpha in osteoblasts, and to study the mechanism of action of Gukan treatment for osteoporosis. Methods Osteoblasts were obtained from newborn rats calvaria by collagenase trypsin digestion. Three groups were included in the experiment: normal serum group, serum of Gukan model group and serum of E2 model group. IL-1 ,IL-6 and TNF-alpha were detected by ELISA. Results Expression of IL-1, IL-6 and TNF- alpha in osteoblasts in the serum of Gukan model group was decreased compared with that in the normal serum group (P〈0.05), but did not show significant difference from those of E2 serum group. Conclusion The serum containing Gukan substances can inhibit the secretion of IL-1, IL-6 and TNF-alpha which may be one of the mechanisms of action of GUKAN treatment for osteoporosis.
