大鼠实验性短暂脑缺血血浆中cAMP相对浓度与血-脑脊液屏障变化的关系

摘要 目的 观察短暂脑缺血再灌流过程中血 -脑脊液屏障超微结构及血浆中环一磷酸腺苷 (c AMP)相对浓度的变化。方法 用改良 pulsinelli法制成短暂脑缺血再灌流模型。用 H3- c AMP放免分析药盒测定血浆中 c AMP相对浓度 ,并观察血 -脑脊液屏障中紧密连接的变化。结果 在短暂脑缺血及再灌流早期 ,血浆中 c AMP相对浓度及紧密连接紧密程度 (闭锁小带长度及凸嵴数目 )无明显变化 ,血管内皮中胞饮小泡数目明显增多。再灌注中期内皮细胞间紧密连接松弛 ,闭锁小带长度及凸嵴数目均减少。血浆中 c AMP浓度亦降低。结论 短暂脑缺血及再灌流早期血 -脑脊液屏障通透性的改变 ,主要是由于内皮细胞中胞饮小泡数目增多所致。随着再灌流时间的延长 ,紧密连接逐渐开放 ,c AMP相对浓度亦随着紧密连接的紧密程度 (闭锁小带长度及凸嵴数目 )下降而下降。因此 ,血浆中c AMP相对浓度可以反映血 Objective To oberserve the change of blood brain barrier ultrastructure and relative concentration of plasma cAMP on experimental transient cerebral ischemia in rat. Methods Using radioimmunoassay technique investigated the relative concentration of cAMP in rat plasma after transient cerebral ischemia and reperfusion,using routine electron microscopy oberserve the change of blood brain barrier tight juction. Results In transient cerebral ischemia and early reperfusion groups,the relative concentration of plasma cAMP didn ,t change significantly,so did the tight junction. But the number of plasma vesicle was more than sham operated groups and early reperfusion groups. With the time of reperfusion,the tight degree of tight junction became looser and looser. At same time,the relative concentration of cAMP in plasma became lower and lower. Conclusion In transient cerebral ischemia and early reperfusion,the reason of blood brain barrier opening was the addtion of plasma vesicle in brain capillary endothelial cell. With the time of reperfusion,the relative concention of cAMP in plasma and the tight degree of tight junction appeared positive correlation.
