摘要
为了探讨新疆杏品种花粉离体培养条件,以新疆3个杏品种库买提、树上干和卡巴克胡安为试材,采用固体培养法研究不同培养基组分、pH值和培养温度对花粉萌发和花粉管生长的影响。结果表明,花粉萌发率和花粉管生长量随蔗糖和硼酸浓度的增加呈先升后降趋势,蔗糖浓度为15%和硼酸浓度为0.01%时出现峰值;随培养温度和培养基pH值的增大也呈先升后降趋势,温度为25℃和pH值为6.0时出现峰值。3个杏品种花粉离体培养的最适培养条件均为:培养基:1%琼脂+15%蔗糖+0.01%硼酸,pH值为6.0,培养温度为25℃。但不同品种花粉生长的适宜培养基组分、pH值和培养温度范围略有差异。
In order to discuss isolated culture condition on pollen of Xinjiang apricot varieties,taking three apricot varieties :Kumaiti,Shushanggan and Kabakehuan in Xinjiang as test materials, using the method of pollen solid sprouting culture, to research the influence of different component parts in culture medium, pH value and culture temperature on pollen germination and pollen tube growth. The result shows that pollen germination rate and pollen tube growth at first increased then reduced with increase of the density of sac- charose and boric acid,the peak value was found when saccharose's density was 15% ,and boric acid's den- sity was 0.01% ;The peak value was also at first increased then reduced with the increase of culture tem- perature and pH value,the peak value of temperature was 25℃ ,and pH value was 6.0. The most suitable culture condition of all the three varieties of apricot's pollen growth was: culture medium:l% agar+15 % saccharose+0.01% boric acid,pH value was 6. 0,cuhure temperature was 25℃. However,there were slight differences in suitable component parts of culture medium, the range of pH value and culture tem- perature among different varieties of pollen growth.
