摘要
以半滑舌鳎为试验对象,对淋巴细胞体外培养和染色体制备条件如促分裂剂、培养时间、培养温度、培养基、秋水仙素浓度及作用时间等进行探索,在建立一套基于半滑舌鳎淋巴细胞体外培养的染色体制备方法。结果表明,对于半滑舌鳎淋巴细胞体外培养,用离心法收集淋巴细胞,培养基为含15%体积分数胎牛血清的L-15培养基,添加3μg.mL-1的PHA-P作促分裂剂,分裂指数可达2.6%±0.2%,6 d后收集细胞制作染色体,获得较好效果。
Half smooth tongue sole（Cynoglossus semilaevis） was used to explore the condition of the peripheral blood lymphocytic cell cultivation and chromosome preparation. The conditions were searched from mitogens, incubation periods, culture temperature, media and the effect of the chloride. The aim was to establish a systemic method. The results showed that lymphocytes isolated from fresh blood by a stirring method were cultured in medium L-15 supplemented with 15% FBS, 3 pg. mL1 PHA （phytohemagglutinin） as mitogens, and collected at day 6. This was the optimized culture condition for half smooth tongue sole. Alter culture to make chromosome, the mitotic index （MI） could reach （2.6±0.2）%.
