活性氧(ROS)对小鼠胚胎分裂指数的影响

摘要 小鼠体内发育的桑椹胚、早期囊胚、囊胚、扩张囊胚和从1-细胞期取出在CZB培养液中培养的桑椹胚、早期囊胚、囊胚、扩张囊胚的分裂指数分别为5.85,4.64,2.16,4.96和6.25,508,2.77,3.03.经统计分析,体内与体外CZB培养的胚胎分裂指数无显著差异,说明体内发育的胚胎和体外CZB培养胚胎的有丝分裂增殖能力没有差异.体外CZB培养的不同时期添加H2O2的胚胎,发育至桑椹胚、早期囊胚、扩张囊胚阶段,制作胚胎标本,胚胎的分裂指数显示:扩张囊胚的0~72h组的分裂指数与0~12h,24~36h组有显著差异(P〈0.05),而与其它各发育阶段的各组分裂指数均无差异,说明经H2O2处理之后存活下来的胚胎与正常体外发育胚胎的有丝分裂增殖能力没有差异. The mitotic indexes of moruia, early blastocyst, blastocyst, expanded blastocyst of mice in vivo were 5.85, 4.64, 2.16,4.96, and the indexes of those developed from 1-cell embryos cultured in CZB media were 6.25, 5.08, 2.77, 3.03 . No significant difference exist- ed in these mitotic indexes of embryos in vivo or in vitro cultured in CZB media by statistic analysis, which indicated that there were no marked differences in mitotic ability to these embryos. The embryos were cultured up to morula, early blastocyst, expanded blastocyst stage in vitro by adding exogenous oxidant-H2O2 to CZB media in different developing periods, then made embryos samples and inspected the divide indexes of those mitotic indexes. The results showed that the mitotic indexes had significant differences( P 〈 0.05) in expanded blastocyst of 0 - 72 hours vs 0- 12 hours treatment group, and 0- 72 hours vs 24- 36 hours treatment group, whereas this differences did not exist in other groups at other different developing stages, which suggested that there was no difference in the mitotic ability between the survived embryos adding H2O2 and normal embryos in vitro .
