摘要
目的:确定白藜芦醇(Resveratrol,Res)对黑素瘤细胞株A375细胞增殖及骨桥蛋白表达的影响。方法:用不同浓度的白藜芦醇处理A375细胞,MTT比色法分别检测白藜芦醇对细胞增殖的抑制作用,RT-PCR和蛋白印迹方法检测骨桥蛋白的mRNA和蛋白的表达。结果:白藜芦醇对A375细胞增殖的抑制呈时间和浓度依赖性;白藜芦醇能显著减少内皮素作用后黑素瘤细胞株A375骨桥蛋白中mRNA和蛋白的表达(P<0.05)。结论:白藜芦醇作为内皮素受体拮抗剂有可能通过抑制内皮素受体(ETR-B)下调骨桥蛋白的表达,进而抑制A375细胞的增殖。
bjective: To investigate the effect of resveratrol (Res) on the proliferation of the human melanoma cells A375 and the gene expression of osteopontin (OPN). Methods: Human melanoma cells A375 were cultured, and was treated with Res in different concentrations. MTr colorimetric assay was used to detect the effect of Res on the proliferation of human melanoma cells and A375, and the protein expression of OPN was detected by RT- PCR and Western blot test respectively. Results: The inhabitation of Res on the proliferation of A375 cells was in a time - and dose - dependent manner. The mRNA and protein expression of OPN were significantly decreased in A375 cells treated with 100μmol/L Res, as compared to the control cells ( P 〈0.05). Conclusion: Res can up- regu- late the expression of OPN via inhibiting ETR- B in A375 cells.
