骨桥蛋白在母胎界面中表达的研究

摘要 目的通过检测骨桥蛋白(osteopontin,OPN)在正常早、中、晚孕期绒毛及蜕膜组织中的定位及表达情况来探讨其在妊娠期的可能作用。方法分别采用免疫组化染色法(SP法)及逆转录-聚合酶链反应(RT-PCR)方法检测早、中、晚孕期绒毛及蜕膜中OPN的表达。结果OPN在正常早、中、晚孕期绒毛及蜕膜中均有表达,其主要定位于细胞滋养层细胞、合体滋养层细胞及蜕膜细胞的胞膜和胞质中,此外在绒毛间质及血管中亦呈阳性表达。OPNmRNA及其蛋白表达水平的比较,在绒毛组织中,早孕期低于中孕期(P<0.05),中孕期低于晚孕期(P<0.05);蜕膜组织中,早孕期的表达量最高,与中、晚孕期比较差异均有统计学意义(均P<0.05),而在中、晚孕期组间比较差异无统计学意义(P>0.05)。结论在胚胎发育进程中,绒毛中OPN的表达量随孕周增加而增加,而蜕膜中以早孕期表达最强,推测它在孕早期可能更多是参与调节侵袭作用,而在中晚孕期可能更多的是维持免疫调节作用。 Objective To detect the osteopontin (OPN) in placenta during pregnancy to explore its role in the maintenance for pregnancy. Methods Immunohistochemical staining (SP method) and the semi-quantitative method of reverse transcription polymerase chain reaction were used to detect the expression of OPN in placenta of normal pregnancy with three periods. Results OPN was expressed in all tissues, and mainly located in membrane and the cytoplasm of the trophoblast cells, syncytiotrophoblast cells and decidua cells. In addition, the expression of OPN in blood vessels and stroma was also positive. In the villi tissue, the protein and mRNA expression of OPN in early pregnancy was lower than the medium-term pregnancy (P〈 0.05), and the expression in the medium-term pregnancy was also lower than that in the late pregnancy (P〈0.05). In the decidua tissue, the expression of OPN in early pregnancy was strongest among three periods of pregnancy (P〈0.05), but there was no significant difference between the medium-term pregnancy and late pregnancy (P〉0.05). Conclusion In the process of embryonic development, the expression of OPN in villi is increased with gestational age, and the strongest expression is detected in decidua in early pregnancy, so we speculate it may be involved in the regulation of invasion in early pregnancy, and may play a role in maintaining immune in the late pregnancy.
