荷丹片对动脉粥样硬化大鼠氧化应激的影响 被引量:6

摘要 目的:观察不同剂量荷丹片对动脉粥样硬化大鼠氧化应激的影响。方法:48只雄性SD大鼠随机分为正常组,模型组,荷丹片高(3g·kg-1·d-1)、中(1.5g·kg-1·d-1)、低(0.5g·kg-1·d-1)剂量组及辛伐他汀组,每组8只。正常组给予普通鼠饲料喂养,其余各组使用高脂饲料造模。12周后取各组大鼠腹主动脉电子显微镜下观察主动脉形态学变化;腹静脉取血测定各组大鼠超氧化物歧化酶(SOD)、丙二醛(MDA)及NO含量。结果:高剂量荷丹片可以升高血清SOD、NO水平,降低血清MDA水平,与模型组相比差异有统计学意义(P<0.05)。荷丹片高剂量组与辛伐他汀组比较差异无统计学意义(P>0.05)。荷丹片组镜下表现主动脉粥样硬化组织损伤明显减轻。结论:高剂量荷丹片通过减轻AS大鼠氧化应激发挥其抗动脉粥样硬化的作用。 Objective: To evaluate the effect of Hedan tablet on oxidant stress in atherosclerosis rats. Methods: Forty-eight healthy male SD rats were randomly divided into normal control group, model group, three Hedan tablet treated groups (3 g. kg-1. d-1, 1.5 g. kg-1. d-1 and 0.5 g. kg-1. d-1) and simvastatin treated group. Rats in normal control group were given ordinary chow, and rats in other groups were given high fat diet to establish the athersclerosis model. After 12 weeks, rats were sacrificed to observe the histopathological changes in abdominal aorta by electronic microscope. The serum levels of superoxide dismutase (SOD), malondialdehyde(MDA)and nitric oxide (NO) were determined in different groups. Results: In comparing with the model group, the serum levels of SOD and NO were significantly increased by Hedan tablet(3 g- kg-1. d-1),and the serum level of MDA significantly decreased(P 〈 0.05). There were no significant differences in levels of SOD,MDA and NO between Hedan tablet treated(3 g. kg-1. d-1)group and simvastatin treated group (P 〈 0.05). The atherosclerosis lesion was significantly reduced in Hedan tablet treated group. Conclusion: Hedan tablets (3 g.kg-1. d-1) can inhibit the atheroselerosis formation in experimental rats.Its mechanism might be related to its anti-oxidation effect.
