摘要 目的 :研究化疗药物氟尿嘧啶 (5 - FU)、丝裂霉素 (MMC)、阿霉素 (Dox)、顺铂 (Cis)对人胃癌细胞株 (SGC)的作用特征 ,探讨 SGC细胞株对单一及联合化疗药物的敏感性 ,化疗药物浓度及作用时间对细胞抑制率的影响。方法 :SGC细胞株在含有不同药物的不同浓度的培养基中孵育不同时间 ,用 MTT法测定细胞的抑制率 ,化疗药物浓度参照其在人体的最高峰值浓度 ,选择其峰值浓度的 1/2 5~ 2倍范围 ,作用时间选择 2~ 2 4h。结果 :SGC细胞株对不同化疗药物的敏感性存在差异 ,对Dox最敏感 ,其次为 5 - FU、MMC,对 Cis不敏感。 5 - FU + MMC、5 - FU + Cis及 5 - FU + Dox+ MMC的联合应用表现了较明显的协同作用 ,其中 5 - FU、Dox及 MMC三药联用最显著 ;尽管 Cis单用不敏感 ,在联合应用中也表现了较强的协同作用。相同药物作用时间下 ,随着药物浓度的增加 ,Dox、5 - FU和 MMC对 SGC细胞株的抑制率明显增加 ,Cis的抑制率无明显变化 ;相同浓度下 ,药物作用时间对 SGC细胞株的抑制率也有显著影响 ;化疗药物的联合应用也表现了相似的特征。结论 :SGC细胞株对不同化疗药物的敏感性不同 ,化疗药物的联合应用可明显提高对细胞株的抑制率 ,单用不敏感的药物 ,联合应用中也可能表现较明显的协同作用 ;在影响药效动力学的主要? AIM: To study the pharmacodynamics of the chemotherapeutic drugs, 5 fluorouracil (5 FU), doxorubicin (Dox), mitomycin (MMC) and cisplatin (Cis) to SGC cell line to explore the sensitivity of the cell line to the single drug and the combined drugs (5 FU+MMC, 5 FU+Cis, 5 FU+Dox+MMC) and the factors that affected the inhibitive rate of the cell line, the concentration of drugs and the time that the cell line contacted with the drugs. METHODS: The SGC cell line was incubated with different concentration of the drugs for different time, referring to the peak concentration in the body, the concentration was from 1/25 to 2 times of the peak concentration, the time was 2 to 24 h. Then the inhibitive rate of cell line was tested with the MTT assay. RESULTS: The difference of sensitivity was found among 4 drugs. Dox was most sensitive, then 5 FU and MMC. The SGC cell line was not sensitive to Cis. The combination of the drugs produced evident synergy, include the Cis. The results also showed, that inhibitive rate of the cell line was increased evidently with the drug concentration increasing as the same time for Dox, 5 FU, MMC and all application of drugs combined, but for Cis, no evident change when the concentration less than the peak concentration. CONCLUSION: The sensitivity of SGC cell line to the different drugs is varied. The combination of the chemotherapeutic drugs can produce the synergy. The drugs that are not sensitive alone can have synergic effect with other sensitive drugs. To SGC cell line, the concentration of drugs and the exposure time affects the inhabitive rate as similar extent.
