不同浓度七氟烷对小鼠学习记忆和海马CA1区及齿状回星形胶质细胞数量的影响

摘要 目的 探讨0.5%、1%、3%三个浓度的七氟烷对小鼠学习记忆和海马CA1区及齿状回GFAP(glial fibrillary acidic protein)阳性细胞数量的影响.方法 将40只小鼠随机分为正常组(未经任何处理) 、0.5%七氟烷组( 0.5%七氟烷吸入麻醉2 h)、1%七氟烷组(1%七氟烷吸入麻醉2 h)和 3%七氟烷组(3%七氟烷吸入麻醉2 h).给予相应的处理后,用 Morris 水迷宫行学习记忆测试.测试完毕后处死小鼠,取脑组织用免疫荧光方法观察海马CA1区及齿状回的星形胶质细胞数量变化.结果 与正常组相比,低浓度七氟烷组(0.5%、1%)小鼠逃避潜伏期无明显改变(P均>0.05),高浓度(3%)七氟烷组小鼠逃避潜伏期缩短,海马CA1区及齿状回GFAP阳性细胞数明显增加(P<0.05).结论 吸入低浓度七氟烷后,小鼠学习记忆无明显变化;吸入高浓度七氟烷后,其学习记忆能力有所增加,可能与其星型胶质细胞数增加有关. Objective To explore the effect of sevoflurane in three concentrations of 0.5% , 1% and 3% on mice learning and memory and the number of GFAP - positive cells in hippocampal CA1 and dentate gyrus. Methods Forty mice were randomly divided into control (without any treatment) , 0.5% sevoflurane group (0.5% sevoflurane anesthesia 2 h) , 1% sevoflurane group ( 1% sevoflurane anesthesia 2 h) and 3% sevoflurane group (3% sevoflurane anesthesia 2 h). Learning and memory test llne was given after the appropriate treatment with the Morris water maze. Then mice were sacrificed followed by observing the change in the number of astrocytes in hippocampal CA1 region and dentate gyrus by immunofluorescence. Results Compared with control group, Low concentration of sevoflurane showed no significant change in Escape latency (P 〉 0.05 ) , High concentrations (3%) of sevoflurane in mice escape latency, hippocampal CA1 and dentate gyms of GFAP - positive cells was significantly increased ( P 〈 0.05 ). Conclusion Anesthesia with low concentrations of sevoflurane, no significant changes in learning and memory in mice; Anesthesia with high concentrations of sevoflurane, its ability to increase learning and memory, may be related to the number of astrocytes increased.
