摘要
用放射性配基结合分析法 ,测定 β 肾上腺素能受体 ( β AR)在大鼠肺微血管内皮细胞 (RPMVEC)的分布情况 ,并观察炎性损伤对它的影响。结果表明 ,β AR的最大结合容量Bmax为 ( 5 .5 83± 0 .30 6 )fmol/1 0 5 细胞 ,Kd值为 ( 0 .2 7± 0 .0 3)nmol/L ;LPS、TNF和H2 O2 作用后 ,β AR的Bmax均较正常对照组显著降低 (P <0 .0 5~ 0 .0 1 )。提示 :RPMVEC胞膜存在高亲和力的 β AR ;LPS、TNF和H2 O2 可引起 β AR的显著下调。
Radioactive ligand binding assay was used to measure the distribution density of beta adrenoceptor(β AR) in rat pulmonary microvascular endothelial cells(RPMVEC) and to observe the effect of inflammatory injury on it. The results showed that the maximum binding capacity (Bmax) of RPMVEC β AR was (5.583±0.306) fmol/10 5cells and the Kd value was (0.27±0.03)nmol/L.The Bmax of β AR significantly decreased compared with the normal control group(P<0.05~0.01). The results suggest that there is a high affinity of β AR in RPMVEC and LPS、TNF as well as H 2O 2 can induce down regulation of β AR individually.
