摘要
目的检测冬凌草甲素对人乳腺癌细胞MDA-MB 231侵袭的影响并探讨其作用的分子机制。方法用Transwell实验检测冬凌草甲素对人乳腺癌细胞MDA-MB 231侵袭的影响;明胶酶谱实验检测冬凌草甲素对基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)酶活性的影响;荧光素酶报告基因实验检测冬凌草甲素对MMP-2和MMP-9启动子活性的影响。结果冬凌草甲素明显抑制MDA-MB 231细胞的侵袭,且明显下调MMP-2和MMP-9的启动子活性和酶活性(P<0.05或<0.01)。结论冬凌草甲素能显著抑制乳腺癌细胞的侵袭,且这种抑制作用可能通过下调MMP-2和MMP-9的启动子活性和酶活性而实现。
Objective To investigate the effects of oridonin on the invasion of human breast cancer cells MDA-MB 231 and the underlying mechanisms. Methods Transwell assay was performed to exam- ine the effects of oridonin on the invasion of human breast cancer cells MDA-MB 231. Gelatin zymography assay was performed to detect the effects of oridonin on the enzyme activities of MMP-2 and MMP-9. Lu- ciferase reporter assay was carried out to test the effects of oridonin on the promoter activities of MMP-2 and MMP-9. Results Oridonin significantly inhibited the invasion of MDA-MB 231 cells. And oridonin treatment led to the decrease of the promoter activities and enzyme activities of MMP-2 and MMP-9 (P 〈 0. 05 or P 〈 0.01 ). Conclusion Oridonin is able to effectively inhibit the invasion of breast cancer cells and this inhibition may be reached through decreasing the promoter activities and enzyme activities of MMP-2 and MMP-9.
