摘要
研究了不同植物激素对药用植物灵菊七愈伤组织诱导、继代培养、细胞染色体稳定性的影响。结果表明,灵菊七的愈伤组织诱导、继代培养分别以MS+4.0 mg/L 6-BA+0.5 mg/L NAA、MS+2.0 mg/L 6-BA+0.5 mg/L NAA为培养基较适宜。培养在MS+2.0 mg/L 6-BA+0.5 mg/L NAA上的愈伤组织细胞中期分裂相染色体数目稳定,为20条,而培养在MS+2.0 mg/L ZT+0.5 mg/L NAA上的愈伤组织细胞中期分裂相染色体数目不稳定,其中86.7%的细胞含有20条染色体,13.3%的细胞含有10条染色体,表明激素ZT会导致愈伤组织细胞的染色体数目不稳定。在81个被观察的愈伤组织细胞中,超过85%的细胞染色体数目为20,表明灵菊七的体细胞染色体数应为2 n=20。
It was investigated that different plant hormones effected on callus induction, callus regeneration, and stability of chromosome number in callus cells of Gyrtura medica. The result showed that medium MS+4.0 mg/L 6-BA+0.5 mg/L NAA, MS+2.0 mg/L 6-BA +0.5 mg/L NAA was fit for callus induction and callus regeneration respectively. Chromosomal number were stable, with 20 chromosomes, when callus was cultured on medium MS+2.0 mg/L 6-BA+0.5 mg/L NAA, while chromosomal number were unstable, when callus was cultured on the medium of MS+2.0 mg/L ZT+0.5 mg/L NAA, and showed 86.7% cells contained 20 chromosomes, 13.3% cells contained 10 chromosomes. That meaned ZT enhanced chromosomes more unstable. In 81 cells , more than 85% callus cells contained stable 20 chromosomes, it was concluded that chromosome number of somatic cell of G^rtura medica should be 2n=20.
