摘要
目的观察锌指转录因子Snail及上皮钙黏蛋白(E.cadherin)在肾透明细胞癌(CCRCC)中的表达,并探讨与CCRCC病理分级、分期和转移等相关生物学行为因素的关系。方法收集我院手术切除的69例CCRCC组织标本,同时取58例癌旁组织和10例正常肾组织标本作为对照,采用免疫组织化学链霉菌抗生物素蛋白.过氧化物酶连结(sP)法检测CCRCC组织、癌旁组织和正常肾组织中Snail和E—cadherin的表达。结果CCRCC组织Snail阳性率(82.7%)显著高于癌旁组织(43.1%,P〈0.01)。CCRCC组织E—cadherin阳性率(31.9%)显著低于癌旁组织(91.5%)(P〈0.01)。两者的表达均与CCRCC不同分化程度、分期、浸润和转移有关(P〈0.05)。CCRCC组织中E—cadherin与Snail的表达呈负相关(r=-0.342,P〈0.05)。结论E—cadherin低表达与Snail高表达可能是CCRCC发生浸润转移的重要生物学标志。联合检测E—cadherin及Snail对预测CCRCC浸润转移有重要意义。
Objective To study the expression of transcriptional factor Snail and adhesion factor E-cadherin in renal clear cell carcinoma (RCCC) and evaluate their correlation with pathological grading, clinical stage, invasion and metastases of RCCC. Methods The expression of Snail and E-cadherin proteins was detected by using immunohistochemical streptavidin-peroxidase method in 69 cases of RCCC tissue, 58 cases of the para-caneerous mucosa and 10 cases of normal renal tissue. Results The positive rate of Snail in RCCC was 82. 7%, which was significantly higher than that in para-cancerous mucosa (43.1% ,P 〈0. 01 ). The positive rate of E-eadherin in RCCC was 31.9% , which was significantly lower than that in para-cancerous mucosa (91.5% ,P 〈 0.01 ). The expression of E-cadherin and Snail was significantly related with the differential degrees, clinical stage, depth of tumor invasion and distant metastasis (P 〈 0. 05 ). There was a negative correlation between the expression of E-eadherin and Snail in RCCC. Conclusion The over-expression of Snail and decreased expression of E-cadherin might be important biological markers for invasion and metastasis of RCCC. The combined detection of E-cadherin and Snail has far-reaching significance on predicting the invasion and metastasis of RCCC.
