摘要
目的探讨黄芪丹参茯苓龟板合剂对肾间质纤维化进展过程的干预作用及其机制。方法通过单侧输尿管梗阻法建立肾间质纤维化的大鼠模型,实验分为假手术(Sham)组、单侧输尿管梗阻(UUO)组、血管紧张素转换酶抑制剂(ACEI)组及黄芪丹参茯苓龟板合剂(中药)组,术后第14天取大鼠肾组织进行常规HE、Masson染色及CD68、α-SMA的免疫组化检测。结果(1)ACEI组及中药组肾间质纤维化程度、CD68和α-SMA的表达均低于UUO组(P<0.05);(2)中药组肾间质纤维化程度、CD68和α-SMA的表达低于ACEI组(P<0.05)。结论黄芪丹参茯苓龟板合剂具有抑制肾间质纤维化的作用,其机制可能是抑制间质单核巨噬细胞的浸润,并阻止间质细胞发生转分化。
Objective To investigate the interventional effect of Chinese herbs on renal tubulointerstitial fibrosis after unilateral urethral obstruction (UUO) in rats. Methods The experimental rats were randomly divided into four groups:sham-operated group for control (Sham group), unilateral ureteral obstruction group (UUO group ), angiotensin converting enzyme inhibitor treatment group (ACEI group) and Chinese herbs group. The kidney tissues were collected from each group on 14d after operation. Hematoxylin-eosin stain and Masson stain were used to measure tubulointerstitial fibrosis and immunohistoehemical method was performed to detect CD68 and or- smooth muscle aetin. Results ( 1 ) The degree of renal tubulointerstitial fibrosis, and the expression of CD68, α-SMA were reduced in ACEI group and Chinese herbs group compared with UUO group( P 〈 0.05 ). ( 2 ) The degree of renal tubulointerstitial fibrosis, the posi- tive rate of CD68 and α-SMA was lower in Chinese herbs group compared with ACEI group ( P 〈 0.05 ). Conclusion Chinese herbs can prevent the renal interstitial fibrosis after unilateral urethral obstruction in rats, through inhibiting the infiltration of mononuclear macro- phage in mesenchymal and transdifferentiation of interstitial cells.
