摘要 目的:观察丹酚酸B对大鼠HgCl2中毒肾间质纤维化模型的作用,并探讨其影响肾脂质过氧化的部分作用机理。方法:雄性大鼠以8mg/kgHgCl2水溶液灌胃9周制备大鼠肾间质纤维化模型。从造模开始SA-B组以丹酚酸B10mg/kg灌胃,VitE组以100mg/kg灌胃,1次/天,共9周。试剂盒方法检测肾功能和脂质过氧化,肾组织HE染色、六胺银染色及Masson染色观察肾组织炎症与胶原纤维沉积变化;盐酸水解法测定肾组织羟脯氨酸(Hyp)含量。结果:丹酚酸B能明显减轻模型大鼠肾小管炎症坏死与肾间质胶原沉积,减轻肾组织Hyp含量,降低异常增高的血清尿素氮与肌酐水平,升高GSH含量,并降低MDA含量。结论:丹酚酸B可明显改善大鼠HgCl2中毒性肾间质纤维化,其机制为增强机体抗氧化损伤能力与减轻脂质过氧化损伤。 Objective:To investigate the effects and mechanisms of salvianolic acid- B (SA- B) against renal interstitial fibrosis in rats induced by administration of mercurto 4 groups: normal group, model group, SA - B treatment group and VitE treatment group. Renal, interstitial, fibrosis model was induced by administration of HgCl2 at a dose of 8mg/kg rat weight every morning for 9 weeks. Meanwhile rats in SA - B treatment group and ic chloride ( HgCl2 ). Methods; The rats were randomly divided inVitE treatment group were treated with SA - B at a dose of 10mg/kg and VitE at a dose of 100mg/kg respectively in the afternoon for 9 weeks. Serum creatinine and urea nitrogen content and renal lipid oxidation injury, namely, activity of superoxide dismutase (SOD) and glutathione peroxidase ( GSH - Px) and content of GSH and malondiahdehyde (MDA) of kidney, were tested with commercial kits. Hydroxyproline (Hyp) content of kidney was assayed with Jamall's method; histological changes of kidney were observed with hematoxylin and eosin (H&E) Stain, masson stain and methenamine silver (PASM) stain. Results: Compared to model group, SA -B improved renal function, decreased kidney Hyp content, alleviated renal interstitial fibrosis, increased GSH content and decreased MDA content in kidney. Conclusions: SA - B could ameliorate renal interstitial fibrosis in rats induced by HgCl2, which was related to increasing organism function against oxidative damage and alleviating lipid oxidation injury.
