摘要
本文以LD50补量法测定中药附子、川乌及四逆汤的体存量,从体存量的经时性变化判断药物在体内的衰减模式并计算表观药动学参数。结果所测药物均符合二室动力学模型,消除相半衰期分别为17.0h.12.1h 及5.8h。
Apparent pharmacokinetic parameters op Radix Aconitum carmichali(Ⅰ),LateralRadix Acouitum carmichali(Ⅱ)preparatious aud“Si Ni Taug”were determinedby acute mortality of mice.For each preparatiou,the iP LD_(50) of mice was deter-mined at first.Mice received an initial dosage(about LD_(10))were divided into 5—6 interval sronps.After appropriate interval the second dosage was siven and ip LD_(50)(t)was determined.Residual of the initial dosage after the interval of twas LD_(50) minus LD_(50)(t).A curve of dynamic change of initial dosage was drawnon the basis of the relationship between interval(t)and residual(Rt),and appar-ent pharmacokinetic parameters were calculated and showed two comportmentmodel.The t1/2β of Ⅰ ane Ⅱ preparations,and“Si Ni Tang”were 17.0h,12.1hand 5.8h respectively.
