摘要
针对柿属植物叶片富含单宁,常规CTAB法提取DNA质量低等问题,为了获得符合柿属植物SSR-PCR扩增要求的DNA,分别采用改良CTAB法和植物基因组DNA提取试剂盒两种方法,提取柿属植物DNA,结果显示:两种方法均可得到满足SSR-PCR扩增要求的DNA;试剂盒较改良CTAB法提取的DNA质量高、杂质少,DNA得率略低,但其操作步骤简单,耗时短,毒害小,因其费用较改良CTAB法高,各实验室可根据自身条件选择适宜方法提取柿属植物DNA。
Tannin is rich in the leaves of Diospyros L.so the quality of DNA extracted by conventional CTAB methord is lower.The DNA from trees of Diospyros L.was extracted by using modified CTAB method and plant genomic DNA Kit in order to compare which method is better.The results show that DNA extacted by the two methods also met demands for ssr-pcr amplification.The quality of DNA obtained by plant genomic DNA Kit was better but the achievement rate of DNA was lower.Moreover,the method of plant genomic DNA Kit was much easier,needed shorter time,had lower toxicity.DNA of trees of Diospyros L.can be extracted by suitable method according to the actual conditions of the lab.
