摘要
用含 50 μmol/ L钇 ( )的氯化钙诱导大肠杆菌进入感受态 ,质粒 p BR32 2的转化率提高2倍 ;若在热休克后使感受态细胞在含 50 μmol/ L氯化钇的营养培养基 (SOC)中复壮 ,则转化率提高 5倍 .实验结果表明 :微量氯化钇存在的条件下 ,钇 ( )可能比钙 ( )更易形成抗 DNase的羟基 -钇 ( ) -磷酸复合物 ,更易于被细胞吸收而提高了质粒的转化率 .
Calcium chloride solution,with a concentration of 50 μmol/L Yttrium(Ⅲ), induces E.coli into competence.The transformation efficiency of plasmid pBR322 will increase by 2 times; If competent cells anabiosis in rich medium with a concentration of 50 μmol/L Yttrium(Ⅲ) after heat shock,the transformation efficiency will increase by 5 times.Experiment results indicate that yttrium(Ⅲ) is easier to form anti\|DNase complex of hydroxyl group\|yttrium(Ⅲ)\|phosphoric acid than calcium(Ⅱ) and yttrium (Ⅲ) enters cell together with complex.
