大剂量维生素C对DNA损伤及槲皮素-7-硫酸酯钠的保护作用 被引量:3

Protective Effect of Sodium Quercetin-7-mono-sulfonate on High-dose Vitamin C Induced DNA Damage

摘要 目的探讨大剂量维生素C(VC)对小牛胸腺DNA损伤及槲皮素-7-硫酸酯钠的保护作用。方法小牛胸腺DNA用VC(0.1、0.25及0.50 mmol/L)预处理和用不同剂量槲皮素、槲皮素-7-硫酸酯钠(0.1、0.25、0.50 mmol/L)与VC(0.50 mmol/L)以不同方式处理(预处理、同时处理),采用琼脂糖凝胶电泳技术检测分析DNA电泳条带及用分光光度法检测分析DNA浓度变化情况。结果低剂量VC组DNA条带及DNA浓度没有明显变化;中高剂量VC组DNA条带明显拖尾,DNA浓度显著升高;各浓度槲皮素-7-硫酸酯钠不同处理组DNA条带拖尾现象较VC损伤组均减弱,小牛胸腺DNA条带变亮,DNA浓度也有明显下降。结论大剂量VC能直接损伤小牛胸腺DNA,槲皮素-7-硫酸酯钠对大剂量VC诱导的DNA损伤有保护作用,且呈明显浓度依赖关系。 Objective To investigate the damage effect of high-dose Vitamin C (VC) on calf thymus DNA and the protective effect of sodium quercetin-7-sulfonate. Methods Calf thymus DNA incubated with VC ( 0. 1,0. 25, 0.50 mmol/L;quercetin and sodium quercetin-7-sulfonate were pretreated or co-treated with VC (0.50 mmol/L) in calf thymus DNA. The changes of DNA were analyzed by agrose gel electrophoresis and spectrophotometry. Results The DNA ladder and DNA concentration had no obvious changing in 0.10 mmol/L VC group,but DNA ladder disappeared and the concentration of DNA obviously increased in 0.25 and 0.50 mmol/L VC groups. Pretreatment or co-treatment with quercetin or sodium quercetin-7-sulfonate,the DNA ladder appeared more clearly and the concentration of DNA decreased obviously. Conclusions These suggest that high-dose VC could induce calf thymus DNA damage and sodium quercetin-7- sulfonate has protective effect on high-dose VC induced DNA damage.
