摘要
目的:研究克林沙星与小牛胸腺DNA之间的相互作用。方法:使用荧光光谱法,根据Stern-Volmer方程及Scatchard方程进行数据处理。采用离子强度的影响、紫外光谱的变化、单双链DNA作用的区别及I-猝灭等实验研究了克林沙星与小牛胸腺DNA间的相互作用模式。结果:在pH值7.0的磷酸盐缓冲液(PBS)中,克林沙星的荧光激发及发射峰分别位于273 nm和427 nm处,小牛胸腺DNA对克林沙星的荧光存在着强烈的猝灭作用,猝灭常数Kq为4.03×104mol-1.L,结合位点数为2.84。结论:克林沙星与小牛胸腺DNA间是一种沟槽作用模式。
Objective: To investigate the interactions between clinafloxacin and calf thymus DNA Methods: The interaction mode was studied by using fluorescence spectrometry. The experimental data were plotted by Stern-Volmer equation and Scatchard equation. Based on ion strength effects, UV spectra, the differences of single stranded DNA from double stranded DNA bound to clinafloxacin and I-quenching effects, the binding interaction mode between clinafloxacin and calf thymus DNA was studied. Results: At pH 7.0 PBS, the excitation and emission wavelength of clinafloxacin were 273 nm and 427 nm, respectively. The addition of calf thymus DNA to clinafloxacin solution resulted in a strong fluorescence quenching with a quenching constant Kq = 4.03 × 10^4mol^-1· L and the binding site size in DNA base pairs n = 2.84. Conclusion: Clinafloxacin interacts with calf thymus DNA in groove bindings.
