摘要
目的:观察丙泊酚对大鼠单肺通气时肺代谢酶的影响,为丙泊酚在胸科麻醉中的应用提供实验研究的数据。方法:40只健康清洁级SD大鼠随机分成四组:P1组:空白组(灌注液中无丙泊酚);P2组:含36mg/L丙泊酚;P3组:含24mg/L丙泊酚;P4组:含12mg/L丙泊酚,每组10只。按随机顺序取大鼠进行实验,称量体重,麻醉,气管切开插管接呼吸机行单肺机械通气1h。然后取出肺组织。-70℃保存,以备统一作Ca2+-ATPase、Na+-k+ATPase测定。结果:肺组织Na+-K+ATPase活力:P1组通气侧活力显著低于P2、P3、P4组通气侧(P<0.01);P1组非通气侧活力显著低于P2、P3、P4组非通气侧(P<0.01)。各组非通气侧均低于同组通气侧(P<0.01或0.05)。肺组织Ca2+-ATPase活力:P1组通气侧显著低于P2、P3、P4组通气侧(P<0.01);P1组非通气侧显著低于P2、P3、P4组非通气侧(P<0.01)。各组非通气侧均显著低于同组通气侧(P<0.01)。结论:通过对大鼠单肺通气时的灌注实验研究发现,丙泊酚对单肺通气时的肺代谢酶有保护作用,其程度与丙泊酚浓度在一定范围内无关。
Objective:To investigate the effect of propofol on metabolic enzyme of rats during one-lung ventilation.Methods:40 rats randomly divided into 4 groups(n=10, each):group P1 (no propofol in the perfusate ), group P2 (the concentration of propofol in perfusate was 36mg/L), group P3(24 nag/L), group P4 (12 rag/L). Metabolic enzyme was quantified after one-lung ventilation .After OLV finished , lung tissue sample were harvest for lhe criterions of the Na^-K+ATPase activity, CaE+ATPase activity.Results:(1)The Na+-K+ATPase activity: as compared to the values in group P2, group P3 and group P4 ventilated lung, the Na+-K+ATPase activity was low in group PI ventilated lung(P〈0.01) ; as compared to the values in group P2, group P3 and group P4airproof lung, the Na+-K+ATPase activity was low ill group PI airproof lung(P〈0.01) ; as compared to the values in reciprocal group ventilated lung, the Na^-K+ATPase activity was low in each group airproof lung(P〈0.01 or 0.05).(2)The Ca2+-ATPase activity: as compared to the values in group P2, group P3 and group P4 ventilated lung, the Ca2+-ATPase activity was low in group Pt ventilated lung(P〈0.01) ; as compared to the values in group P2 , group P3 and group P4 airproof lung, the Ca2+-ATPase activity was low in group Pl airproof lung(P〈0.01) ; as compared to the values in reciprocal group ventilated lung, the Ca2+-ATPase activity was low in each group airproof lung(P〈0.01).Conclusion:These results indicate that the propofol show the protective effects on lung metabolic enzyme of rats during one-lung ventilation , and the effects do not seem to correlate with the concentration of propofol.
