摘要 目的采用MDCK细胞模型研究广防己的肾毒性,探讨MDCK细胞模型作为体外评价中药肾毒性方法的可行性。方法选择马兜铃和苦参作为阳性对照与阴性对照,通过MTT法测定细胞存活率,倒置显微镜观察细胞形态及乳酸脱氢酶(LDH)法检测细胞膜损伤,研究广防己对MDCK细胞的毒性作用。结果给药24 h后,各浓度的广防己组与阳性对照马兜铃组的细胞存活均被明显抑制,且倒置显微镜下可见细胞形态收缩变圆,部分细胞破裂脱落。通过比较乳酸脱氢酶的释放率,马兜铃与广防己对MDCK细胞膜的损伤明显高于阴性对照苦参组,统计学分析结果具有显著性差异(P<0.01)。结论 MDCK细胞模型可用于初步评价中药的体外肾毒性。 Objective To evaluate the nephrotoxicity induced by Chinese herbal with MDCK cells and investigate the possibility of MDCK cells used to assess the nephrotoxicity in vitro.Methods MTT assay,cells morphology and lactate dehydrogenase(LDH) release test were applied to examine the cytotoxicity.Result MDCK cells treated by aristolochia debilis and fangchi root 24 h later were obviously inhibited,and the MDCK cells were contracted and rounded.The LDH release rates were much higher compared with sophora flavescens.Conclusion MDCK cells have different responses to aristolochia debilis,fanschi root and sophora flavescens in terms of nephrotoxicity.MDCK cells could be used to assess the nephrotoxicity of traditional Chinese medicine in vitro
