摘要
目的检测MMP-9和Ezrin在不同类型子宫内膜中的表达情况及与雌、孕激素受体的相关性,探讨其在子宫内膜癌发生、发展中的意义。方法采用组织芯片技术及免疫组织化学方法检测和分析MMP-9和Ezrin在子宫内膜癌43例、子宫内膜不典型增生18例、单纯性增生及复杂性增生20例及正常增生期子宫内膜20例中的表达情况及与雌、孕激素受体的相关性。结果 MMP-9和Ezrin在正常增生期子宫内膜,单纯、复杂性增生,不典型增生及子宫内膜癌中的阳性率呈上升趋势,差异有统计学意义(P<0.01)。MMP-9表达强度在子宫内膜癌FIGO分期之间、浅深肌层浸润组间及有无淋巴结转移组间差异有统计学意义。Ezrin表达强度在子宫内膜癌浅深肌层浸润组间和组织类型间差异有统计学意义。MMP-9与Ezrin在子宫内膜癌上表达呈正相关(P<0.05),而Ezrin与ER在子宫内膜癌上表达呈负相关(P<0.05)。结论 MMP-9与Ezrin在子宫内膜癌的发生、浸润转移中起促进作用,Ezrin可能参与了雌激素受体阴性肿瘤的扩散和转移。
Objective To detect the expression of MMP-9 and Ezrin in different kind of endometrial membranes tissues and the relation with the receptors of estrogen and progestin and to elucidate the role of them in the pathogenesis mechanism of endometrial carcinoma.Methods Tissue microarray and immunohistochemical staining were used to detect the expression of MMP-9 and Ezrin in 43 cases of uterine endometrial carcinoma,18 of atypical endometrial hyperplasias,20 of complex endometrial hyperplasias and 20 of normal endometrium and the relation with the receptors of estrogen and progestin.Results The positive expression rates of MMP-9 and Ezrin in normal endometrium during proliferative phase,simple hyperplasia,complex hyperplasia,atypical hyperplasia and endometrial carcinoma were rising.The difference was statistically significant(P0.01).The difference of expression intensity of MMP-9 was statistically significant in the FIGO operation stage,shallow and deep muscular layer invasion group,and lymphatic metastasis groups.The difference of expression intensity of Ezrin was statistically significant in shallow and deep muscular layer invasion group,and histologic type groups.The relationship between MMP-9 and Ezrin in endometrial carcinoma was correlated positively(P0.05).The expression of Ezrin in endometrial carcinoma was significantly correlated negatively with ER(P0.05).Conclusions MMP-9 and Ezrin play an important role in the process of the occurrence,development,invasion and metastasis of the endometrial carcinoma.Ezrin may participate in the invasion and metastasis of estrogen receptor-negative tumors.
