摘要
[目的]探讨乌药不同溶剂提取物的抑菌和杀菌活性,为其合理开发提供依据。[方法]采用浓度70%乙醇、浓度60%丙酮及沸水提取乌药的活性成分,以金黄色葡萄球菌、大肠杆菌、绿脓杆菌和肺炎克氏菌为供试菌,通过纸片琼脂扩散法测定抑菌圈大小、平板2倍稀释法统计最低抑菌浓度(MIC)和试管2倍稀释法统计最低杀菌浓度(MBC)。[结果]乌药的3种溶剂提取物对4种供试菌均有一定的抑制作用,其中浓度60%丙酮和浓度70%乙醇提取物的抑制效果优于沸水提取物。浓度60%丙酮提取物对金黄色葡萄球菌的抑制效果最明显,抑菌圈大小为(12.08±1.01)mm,MIC和MBC分别为50和125 mg/ml;而浓度70%乙醇提取物对肺炎克氏菌的抑杀效果最好,抑菌圈大小为(11.41±0.65)mm,MIC和MBC分别为200和250 mg/ml。[结论]乌药不同溶剂提取物具有明显的抑菌杀菌作用,尤其是浓度60%丙酮提取物和浓度70%乙醇提取物。
[ Objective ] The aim was to study the bacteriostatic and bactericidal activity of different solvent extracts from RADIX LINDERAE and provide references for its reasonable development. [ Method ] RADIX LINDERAE was extracted with 70% ethanol ,60% acetone and boiled wa- ter. Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa and Klebsiella pneumoniae were used as the experimental strains. The diame- ters of inhibition zone were determined by agar plate diffusion method. The minimal inhibition concentration (MIC) was calculated using agar doubling dilution method. The minimal bactericidal concentration (MBC) was measured by tube doubling dilution method. [ Result ] The three ex- tracts all had inhibitive effects on four strains. The inhibitive effects of the 60% acetone extracts and 70% ethanol extracts were better than that of boiled water extracts. The bacteriostatic activity on Staphylococcus aureus of the extracts which treated with 60% acetone was the best ,the diam- eter of inhibition zone, MIC, MBC was (12.08 + 1. O1 ) mm ,50 mg/ml and 125 mg/ml,respectively; and the anti-bacterial and bactericidal activ- ity on Klebsiella pneumoniae of the extracts treated with 70% ethanol was the best, the diameter of anti-bacterial zone, MIC and MBC was ( 11.41 + O. 65 )ram,200 mg/ml and 250 mg/ml ,respectively. [ Conclusion ] Obvious anti-bacterial activity was observed in RADIX LINDERAE extracts, especiallv that treated with 60% acetone and 70% ethanol.
