摘要
为建立陕西卫矛高效再生体系,以幼茎为外植体诱导愈伤组织,在成功继代愈伤组织的基础上,进行愈伤组织再分化培养研究。采用正交设计方法,考察了6-BA、NAA及Ca2+3个因素对陕西卫矛愈伤组织再分化率的影响。结果表明:再分化培养基中添加3mg/L 6-BA+0.3mg/L NAA+5mmol/L Ca2+,能明显提高陕西卫矛愈伤组织的再分化率。
In order to establish a high efficient regeneration system,the stems of E.schensianus were taken as explants and induced callus formation after having plenty proliferated calli successfully,and then the calli were placed on different combinations regeneration medium.An orthogonal test was carried out to observe the influences of the 6-BA,NAA and Ca2+ concentration on regeneration rate.The results showed that the suitable regeneration medium was MS containing 3.0 mg/L 6-BA,0.3 mg/L NAA and 5.0 mmol/L Ca2+,it could obviously improve the frequency of regenerated shoots.
