摘要
采用正交实验法优选枣核中总黄酮的最佳超声提取工艺。以芦丁为对照品,采用分光光度法进行测定,以枣核中黄酮含量作为考查指标,通过单因素实验研究超声波功率、提取时间、提取温度、料液比和提取溶剂乙醇体积分数对提取效果的影响,再利用正交实验法优化最佳提取工艺条件。结果表明,对枣核黄酮提取影响的大小次序为提取温度>料液比>提取时间>乙醇体积分数>超声波功率,最佳提取工艺为:提取温度55℃,料液比为1:50,提取时间为40min,乙醇体积分数为50%,超声波功率为400W,在该条件下枣核黄酮粗品的提取率为12.45%,粗品中黄酮含量为6.83mg.g-1,能显著提高枣核黄酮的提取效率。
The optimal condition for ultrasonic extraction of flavone from seeds of Chinese dates was investigated by orthogonal test.The total flavone content of seeds of Chinese dates was determined by using spectrophotometry method with rutin as reference substance.In the single factor analysis,the effect of ultrasonic-wave power,extraction time,extraction temperature,solid-liquid ratio and ethanol(extraction solvent)volume fraction were measured.The orthogonal test was used to optimize the conditions of extraction.The results showed that the order of extraction effectiveness was as follows:extraction temperaturesolid-liquid ratioextraction timeethanol volume fractionultrasonic-wave power.The optimum conditions were extraction temperature 55℃,solid-liquid ratio1:50,extraction time 40min,ethanol volume fraction 50%,and ultrasonic-wave power 400W.Under the optimized extraction conditions,the yield of flavone crude product was 12.45%,the content of total flavone was 6.83mg·g-1.The results indicated that operating efficiency of extracting total flavone from seeds of Chinese dates was remarkably increased by orthogonal test and ultrasonic wave extraction.
