摘要 目的检测受体介导基因转移人肿瘤坏死因子α基因的靶向性和表达效率。方法人转铁蛋白同多聚赖氨酸共价交联,盐键合成交联蛋白和外源基因质粒的超分子复合物,对表达转铁蛋白受体的肿瘤细胞株进行β-半乳糖苷酶和人肿瘤环死因子α基因转移。结果检测到外源基因表达;转染人肿瘤坏死因子α基因后细胞上清液检测到一过性人肿瘤坏死因子α活性升高。结论受体介导基因转移具有良好的靶向性,但表达效率低。 Objective To investigate the target and expression efficiency of receptor -mediated TNFα gene delivery. Methods A molecular conjugate vector containing human transferrin was formed and complexed to plasmid pSVβ-gal or pSVk3-TNFαs. The complex was introduced into tumor cell lines which presented different quantity human transferrin receptor. Results The cells could secrete TNFα transiently after being transfected, which could be verified by the L929 cell-killed test and ELISA, as well as a series of molecular biologic methods. Conclusion More special target and lower efficiency were found in receptor-mediated gene transfer than in liposome mediated gene transfer.
