移居高原地区青年男性细胞遗传学损伤的观察分析

摘要 目的研究高原环境下长期作业对移居青年男性细胞遗传学损伤的影响。方法采用自身前后对照的方法,选择驻守高原的某部官兵,利用外周血淋巴细胞培养技术、姐妹染色单体交换(SCE)和微核生成分析技术,对移居高原地区青年男性的细胞遗传学损伤指标进行观察分析。结果观察组驻守高原前微核细胞率和微核率分别为(0.41±0.03)和(0.49±0.03),与对照组的(0.43±0.06)和(0.53±0.07)相比无显著差异(P>0.05);驻守高原一年后观察组的微核细胞率和微核率有明显升高,分别为(0.60±0.05)和(0.76±0.07),与一年前相比有显著性差异(P<0.05)。SCE频率观察组驻守高原前为(6.25±1.24)/细胞,分布范围为(4.1~8.9)/细胞;对照组为(6.61±1.29)/细胞,分布范围在(4.2~8.7)/细胞,两组间无显著性差异(P<0.05)。驻守高原一年后观察组SCE频率为(8.93±1.52)/细胞,分布范围为(4.9~10.6)/细胞,与一年前相比有显著性差异(P<0.05)。结论高原环境对移居人群具有一定的遗传损伤作用,长期在高原环境作业可导致染色体断裂和DNA损伤,其详细机制仍待进一步研究。 Objective To investigate whether it has cytogenetic damage to the men working in plateau area for a long term.Methods To make a prevalence survey and own control study on the officers and soldiers serving in the army who work in plateau area for a long term,and to detect the cytogenetic damage by Peripheral blood lymphocyte culture?technique,sister chromatid exchange(SCE)and micronucleus analysis.Results The micronucleus cell rate and micronucleus rates of the observation group were(0.41±0.03) and(0.49±0.03) before they stationed at high altitude,no significant difference with the control group’s(0.43±0.06)and(0.53±0.07)(P〉0.05).And all these parameters get significantly higher after be in plateau for one year,were(0.60±0.05)and(0.76±0.07) respectively(P〈0.05).The SCE frequency of the observation group were(6.25±1.24)/cell before they stationed at high altitude,and the distribution range were(4.1-8.9)/cell,no significant difference with the control group’s(6.61±1.29)/cell,same as the distribution range(4.2-8.7) /cell(P〈0.05).After stationed at high altitude for one year,the observation group have the SCE frequency(8.93±1.52)/cell,and the distribution range(4.9-10.6) /cell,compared with a year ago,there was significant difference(P〈0.05).Conclusion High altitude may have some of the genetic damage to people who moved to high altitude,long-term operation in high altitude can lead to chromosome breakage and DNA damage,but its detailed mechanism remains to further study.
