摘要
【目的】建立高效液相色谱法测定醋酸奥曲肽中奥曲肽含量的方法。【方法】色谱柱:ZORBAX 300SB-C8柱(2.1 mm×150 mm,5μm),流动相:A:0.1%TFA水B:0.1%TFA乙腈,流速:0.25 ml/min,柱温:40℃,检测波长:210 nm,进样量:20μl。【结果】奥曲肽浓度在0.08~0.2 mg/ml范围内,线性关系良好,r=0.9999;检测限为0.02 mg/ml;加样回收率平均为101.28%,RSD为0.58%。【结论】分析方法可行,准确度高,此法可用于醋酸奥曲肽中奥曲肽含量的测定。
【Objective】To establish a HPLC method for the determination of the content of octreotide in octreotide acetate.【Methods】ZORBAX 300SB-C8 column(2.1mm×150 mm,5μm) and 0.1%TFA water 0.1%TFA acetonitrile as the mobile phase at a flow rate of 0.25ml/min were used with a column temperature of 40℃,a detection wavelength of 210 nm and an injection volume of 20 μl.【Results】The standard curve of octreotide was linear in the range of 0.08~0.20 mg/ml,r=0.9999.The detection limit was 0.02 mg /ml.The average recovery rate was 101.28%,and RSD was 0.58%.【Conclusion】The method is practicable and accurate and can be used for the determination of the content of octreotide in octreotide acetate.
