摘要
【目的】探讨MicroRNA-34c(miR-34c)在膀胱移行细胞癌中的表达及其在膀胱癌发生、发展中的意义。【方法】运用荧光定量real-time PCR检测miR-34c在膀胱癌组织中及癌周非瘤组织的表达水平,采用2-△△Ct算法计算膀胱癌组织与癌周非瘤组织中miR-34c表达量的比值,结合术后病理参数,运用统计学方法分析miR-34c与临床病理资料之间的相关性。【结果】在人膀胱癌组织中发现miR-34c较其相应周围非瘤组织明显低表达。在26例膀胱癌组织中,16例(61.9%)miR-34c显著低表达。miR-34c的低表达与肿瘤大小和恶性程度显著相关。肿瘤越大、恶性程度越高,miR-34c的表达量越低。【结论】低表达miR-34c可能是膀胱移行细胞癌组织的重要特征,并参与膀胱癌的生物学进程,可能对膀胱癌的诊断及防治提供新的思路。
【Objective】 To investigate the expression and clinical significance of microRNA-34c(miR-34c)in bladder transitional cell cancer.【Methods】We detected the relative expression of miR-34c in bladder cancer tissues of 26 patients and compared with that of the matched non-tumor adjacent tissues(NATs) by RNA isolation,Poly(A) tail and real-time RT-PCR.Data analysis was performed with relatively quantitative 2-△△Ct method.We also analyzed the association between miR-34c expression and clinical pathological chara-cteristics in bladder cancer by Chi-Square Test.【Results】We found that the significant low-expression of miR-34c in bladder cancer tissues relative to their NATs.Among 26 patients with bladder cancer,16(61.9%) cases showed a significant low-expression of miR-34c.Low expression of miR-34c was associated with increased tumor size and malignant degree.The patients with low expression of miR-34c tended to have larger tumor size and higher malignanly degree of.【Conclusion】The relative low expression of miRNA-34c may be involved in the tumorigenesis and development of bladder cancer.It may provide a new idea of diagnosis,prevention and treatment of bladder cancer.
