Expressions of matrix metalloproteinase-7 and tissue inhibitors of metalloproteinase-3 in human giant cell tumor of bone and its clinical significancer

摘要 目的探讨基质金属蛋白酶7(MMP-7)及基质金属蛋白酶抑制剂3(TIMP-3)在骨巨细胞瘤中的表达及临床意义。方法 38例骨巨细胞瘤标本,通过免疫组化染色方法检测骨巨细胞瘤标本中的MMP-7、TIMP-3和CD34的表达,观察其在非复发肿瘤组和复发肿瘤组中的变化。分组统计学采用t检验、χ2检验和spearman等级相关分析。结果在骨巨细胞瘤中,MMP-7和TIMP-3阳性率分别为97.0%和95.0%。MMP-7的表达和MVD在复发组和非复发组之间的差异有统计学意义(P<0.05),MMP-7与微血管密度(MVD)存在正相关性(P<0.05),但TIMP-3的表达在复发组和非复发组之间的差异无统计学意义(P>0.05),MMP-7和组织学分级(spearman等级相关分析,P>0.05)不存在相关意义,TIMP-3、MVD和组织学分级(spearman等级相关分析,P<0.05)存在相关意义。结论 MMP-7和TIMP-3在骨巨细胞瘤中高度表达,MMP-7可能会加速肿瘤血管形成、细胞增殖并影响骨巨细胞瘤的复发。 Objective To study the expressions of Matrix Metalloproteinase-7 and Tissue Inhibitors of Metalloproteinase-3 in Human Giant Cell Tumor of bone and its Clinical significance.Methods 38 cases of bone giant cell tumor were slected to study.The expression of MMP-7,TIMP-3,CD34 in Giant cell tumors of bone and its changes in non-recurrent tumor group and the recurrent tumor group were observed by immunohistochemistry.The results of observation was tested by t test,χ2 test and spearman rank correlation analysis.Results The esults showed that positive rates of MMP-7 and TIMP-3 were 97.0% and 95.0% nespectively.The statislical nesults had significant difference(P0.05) between MMP-7 expression and MVD in the recurrence group and the non-recurrence group.MMP-7 correlated positively with MVD(P0.05),but no significant difference in TIMP-3 expression between these two groups(P 0.05);there was no correlation significance between MMP-7 and histological grade(spearman rank correlation analysis,P 0.05) and had correlation in TIMP-3,MVD and histological grade(spearman rank correlation analysis,P0.05).Conclusion MMP-7 and TIMP-3 were both highly expressed in the giant cell tumor of bone.MMP-7 may promote the tumor angiogenesis,cell proliferation and affect the recurrence of giant cell tumor of bone.
