摘要
以白皮松成熟胚的子叶作为外植体进行组织培养,利用秋水仙素诱导多倍体。将子叶预培养10 d后,用质量体积分数为0.01%、0.03%、0.05%的秋水仙素分别浸泡6 h、12 h、24 h,观察子叶生长状态及芽分化状况,20 d后鉴定染色体加倍情况。结果表明:不同浓度的秋水仙素均对子叶生长状态产生影响,浓度越高、处理时间越长,死亡率越高;秋水仙素处理过的子叶芽再生缓慢,且芽再生率随浓度增高而降低;秋水仙素处理后的子叶染色体产生加倍,但随着秋水仙素浓度的提高和处理时间的延长,多倍体诱导率呈现出下降的趋势。数据显示质量分数为0.03%的秋水仙素处理12 h的组合,多倍体诱导率最高,为40.5%。本研究初步建立了白皮松多倍体诱导体系。
Cotyledon explants of mature embryos of Pinus bungeana tissue were culture,the use of colchine induced polyploid.Cotyledons were cultured for 10 days,with the quality of volume fraction of 0.01%,0.03%,0.05% of colchicine immersion,respectively 6 h,12 h,24 h,observation of cotyledon growth and bud differentiation status,identification of chromosome doubling in 20 days.Results indicated that:different concentrations of colchicine effectfed cotyledon growth effect.Concertration was higher,processing time was longer,the higher the death;colchicine treated shoot regeneration is slow,and shoot regeneration rate increased with the concentration decreased;colchicine treated cotyledons chromosomes have doubled,but with colchicine concentration enhancement and treatment the extension of time,polyploid induction rate showed a downward trend.Data displayed quality score of 0.03% for the combination of 12 h treated with colchicine,polyploid induction rate was highest,40.5%.The study on polyploid induction of Pinus bungeana system were initially established.
