摘要
目的 :观察低氧时大鼠肺动脉平滑肌细胞 (PASMC)受血管舒张因子一氧化碳的作用。方法 :随机分为常氧组、低氧组 (1%O2 + 5 %CO2 + 94%N2 )、一氧化碳组 (3 %CO + 5 %CO2 + 92 %N2 ) ,采用流式细胞仪、增殖细胞核抗原 (PCNA)、[3H]-胸腺嘧啶 ([3H]-TdR)摄取量 ,检测PASMC增生的情况。结果 :(1)流式细胞仪检测低氧组PASMCG2 /M期细胞比例明显增多 (P <0 0 1) ,G0 /G1期细胞比例明显减少 (P <0 0 1) ,CO组与常氧组的PASMC各期细胞数差异不显著 (P >0 0 5 )。 (2 )PCNA免疫组化染色 ,低氧组PASMC细胞核阳性反应颗粒明显增加 ,而CO组为弱阳性。 (3) [3H]-TdR的摄取量 ,低氧组可明显促进PASMC摄取 [3H]-TdR(P <0 0 5 ) ,CO组与常氧组比没有明显差异 (P >0 0 5 )。结论 :低氧可以促进PASMC的增殖 ,而CO可以部分抑制低氧对PASMC的作用。
AIM:To explore the effect of carbon monoxide (CO) on hypoxic proliferation of rat pulmonary arterial smooth muscle cell(PASMC). METHODS:Rats were randomly divided into four groups, including: (1)N:Normoxia group (21% O 2); (2)H:Hypoxia group (1% O 2); (3)CO+H:CO+hypoxia group (3% CO+1% O 2),tested by immunocytochemical analysis,[3H]-TdR incorporation and flow cytometry (FCM). RESULTS: (1)Flow cytometric DNA analysis revealed that hypoxia induced significantly enhancement of G 2/M phase and decreased G 0/G 1 phase of PASMC ( P <0.01).The quantities of G 2/M phase or G 0/G 1 phase in CO+H was about equal to that in N. (2) Immunocytochemical reaction of proliferating cell nuclear antigen (PCNA) in H of PASMC was more stronger than that in N or CO+H. The reaction of PASMC in CO+H was similar to that in N. (3)[3H]-TdR incorporation was increased gradually at the end of 12 h in hypoxia and then significanlty promoted at the end of 24 h in hypoxia.But [3H]-TdR incorporation in CO+H was not much enhancedt. (4)α-actin and muscle fiber was decreased in H, but wasn't changed in CO+H. CONCLUSION: Hypoxia may stimulate PASMC and promote proliferation of PASMC indirectly, and low density(3%) carbon monoxide can partly offset the action of hypoxia effectively.
