姜黄素对佐剂性关节炎大鼠滑膜血管新生和低氧诱导因子1α表达的影响 被引量:4

摘要 目的观察佐剂性关节炎大鼠滑膜血管新生和低氧诱导因子1α(HIF-1α)的表达及姜黄素的影响,探讨姜黄素治疗类风湿关节炎可能的机制。方法取SD大鼠,制成佐剂性关节炎模型,设正常组、模型组、姜黄素组、甲氨蝶呤组。造模后9天起分别给予腹腔注射姜黄素50mg.kg-1.d-1,连续14天;甲氨蝶呤0.5mg.k-1.d-1,连续5次。取膝关节滑膜,行免疫组织化学染色,观察滑膜微血管密度(MVD),半定量计算HIF-1α表达。结果①模型组大鼠滑膜MVD(11.10±1.50)分和HIF-1α(1.50±0.33)分表达较正常组增加明显(P<0.01);②治疗后甲氨蝶呤组的MVD(8.73±1.15)分,HIF-1α(0.85±0.34)分和姜黄素组的MVD(8.81±1.14)分,HIF-1α(0.80±0.26)分与模型组比较差异均有统计学意义(P<0.01),且甲氨蝶呤组、姜黄素组组间比较差异无统计学意义(P>0.05)。结论姜黄素能有效抑制佐剂性关节炎大鼠滑膜血管新生和HIF-1α的表达,这可能是姜黄素治疗类风湿关节炎的作用机制之一。 Objective To explore the effect of curcumin on angiogenesis and hypoxia-inducible factor 1-α(HIF-1α) expression in synovium of adjuvant arthritis rats for the mechanism of curcumin treatment of rheumatoid arthritis.Methods Forty SD rats were divided into normal control group,model group,curcumin group and methotrexat group,modeling with complete Freund adjuvant.The rats of curcumin group were treated with curcumin intraperitoneal injection 50 mg·kg-1·d-1 continuously for 14 days and rats of methotrexat group were treated with methotrexat intraperitoneal injection 0.5 mg·kg-1·3 d-1 for 5 times.Synovium micro vessels density(MVD) were observed and the expression of HIF-1α was detected by immunohistochemical test.Results ①In model group,MVD(11.10±1.50) scores and HIF-1α(1.50±0.33) scores showed higher expression than normal control group(P0.01);②After treatment,immunohistochemical test showed that the expression of MVD,HIF-1α all had significant difference compared curcumin group MVD(8.80±1.14) scores,HIF-1α(0.80±0.26) scores and methotrexat group MVD(8.73±1.15) scores,HIF-1α(0.85±0.34) scores to model group(P0.01),and the difference between curcumin group and methotrexat group were not significant(P0.05).Conclusion Curcumin can inhibit HIF1-α expression in the synovium,and reduce MVD.It might be one of the machanisms of curcumin curing rheumatoid arthritis.
