Rg_1对失血性休克大鼠肠上皮细胞PGC-1基因表达的影响

摘要 目的探讨Rg1对失血性休克大鼠肠上皮细胞PGC-1基因表达的影响,以从基因调控水平探讨Rg1对失血性休克时肠上皮细胞线粒体缺血缺氧性损伤的保护作用。方法用RT-PCR方法观察失血性休克大鼠肠上皮细胞PGC-1基因mRNA量的改变,用Western-blot法检测肠上皮细胞PGC-1蛋白的表达变化。结果失血性休克5 hPGC-1 mRNA表达明显降低(P<0.05),复苏后3 h组其表达量仍低于休克前水平,但较失血性休克组表达有所增强。复苏加Rg1治疗3 h后PGC-1 mRNA明显呈较高水平表达。复苏加SB202190及复苏加茴香霉素对PGC-1mRNA几乎无调节作用;Rg1在SB202190存在时,仍微弱上调PGC-1 mRNA表达,Rg1能增强茴香霉素上调PGC-1mRNA的作用。失血性休克5 h大鼠肠上皮细胞PGC-1蛋白表达量明显降低,复苏加Rg1能上调PGC-1蛋白表达,复苏加SB202190或茴香霉素对PGC-1蛋白表达调节作用较弱。Rg1在SB202190存在时,微弱上调PGC-1蛋白表达,但Rg1和茴香霉素同时作用时蛋白表达量明显增加。结论复苏加Rg1可明显上调PGC-1 mRNA及其蛋白表达。提示PGC-1 mRNA上调可以促进PGC-1蛋白表达,PGC-1蛋白合成在转录后水平的调节非常重要。 Objective To study the effect of panax notoginseng saponins Rg1(Rg1) on expression of peroxisome proliferator-activated receptor γ coactivator-1 gene(PGC-1 gene) of small intestional epithelia in rats with hemorrhagic shock,and to investigat protective mechanism of Rg1 on the mitochondrial damage induced by ischemia-hypoxia after hemorrhagic shock on the level of gene modulation.Methods The expressions of intestinal PGC-1 gene were measured by means of RT-PCR.The changes of expressions of PGC-1 protein were examined by Western-blot.Results The RT-PCR results showed that the expression of PGC-1 mRNA lowered significantly in hemorragic shock 5 h group compared with control group(P 0.05).In resuscitation 3 h group,the expression of PGC-1 mRNA increased slightly compared with hemorrhagic shock group,but its level was lower than that of its expression before hemorrhagic shock(P 0.05).SB202190 or anisomysin regulated hardly the expression of PGC-1 mRNA.Rg1 up-regulated slightly and obviously PGC-1 protein expression at the presence of SB202190 and anisomysin respectively.The expression of PGC-1 protein lowered markedly in hemorragic shock 5 h group compared with control group.The expression of PGC-1 protein increased markedly after treatment by Rg1 and anisomysin compared with resuscitation group(P 0.01).Effect of SB202190 and anisomysin on expression of PGC-1 protein of small intestinal epithelial cells in hemorrhagic shock rats were small.Rg1 up-regulated slightly and obviously PGC-1 protein expression at the presence of SB202190 and anisomysin respectively.Conclusion Rg1 up-regulates obviously expression of PGC-1 mRNA and its protein.It is very important to regulate PGC-1 gene expression on the transcriptional and post-transcriptional level.
