20(S)-原人参二醇抑制小细胞肺癌A549细胞增殖的研究 被引量:1

摘要 采用台盼蓝和MTT法测定20(S)-原人参二醇(Ppd)对肺癌A549细胞增殖的抑制作用,Hochest 33258染色和DNA Ladder方法观察Ppd对小细胞肺癌A549细胞凋亡及周期的影响。结果表明,Ppd对肺癌A549细胞具有明显的抑制作用并有剂量时间依赖关系,可明显降低肺癌A549细胞的存活率和活性(IC50为20.7μmol/L);染色后肺癌A549细胞核出现典型的凋亡形态学变化,与DNA Ladder电泳条带弥散情况一致。因此,Ppd对肺癌A549细胞的增殖有明显的抑制作用。 To study the effects of 20(S)-protopanaxadiol (Ppd)on the proliferation of human lung carcinoma A549 cell line. Human lung carcinoma A549 cell was treated with Ppd ,inhibition of Ppd on proliferation of lung cancer A549 cells was determined with trypan blne and MTT, cell grow thrate was determined with MTT assay. The effects of Ppd on A549 cell and cell cycle were observed by the flow cytometer. Results showed that Ppd could inhibit the prolife ration of human lung carcinoma A549 cell in atime and dose dependent manner. Ppd inhibited the growth of human lung carcino- ma A549 cells when IC50 was 20.7μmol/L. The morphological changes were observed in the nuclei of human lung carcinoma A549 cells treated by Ppd. The DNA Ladders bands were detected.Therefore,Ppd could inhibit the proliferation of lung cancer A549 cell.
