摘要
目的:运用反相高效液相色谱法(RP-HPLC)检测丹参五味子片中丹参酮ⅡA、五味子甲素的含量。方法:色谱柱选择Phenomenex KinetexC18(250mm×4.6mm,5μm),以甲醇-水为流动相,甲醇∶水为85∶15,流速设定为每分钟1ml。丹参酮ⅡA的检测波长为270nm,五味子甲素的检测波长为254nm;色谱柱温度设定为28℃;每次进样量为25μg;色谱柱量程为1.0AuFS。结果:丹参酮ⅡA和五味子甲素的线性范围分别为0.3232~1.616μg与0.525~2.625μg,平均回收率分别是99.08%、98.37%,RSD测定值分别为0.42%、0.73%。结论:运用反相高效液相色谱法可准确快速地对丹参五味子片中丹参酮ⅡA及五味子甲素的含量进行测定,操作简便,重复性好。
Objective:To set up a determination of tanshinoneⅡA and deoxyschizaadrin in danshenwuweizi tablet by RP-HPLC.Methods:Two components were separated through Kromasil C18(250 mm×4.6 mm,5 μm)column with methanol-water(85:15)by volume as a mobile phase.The flow rate was 1 ml/min and the detection wavelength were 270 nm(tanshinoneⅡA)and 254 nm(deoxyschizandrin).Results:The linear range of tanshinoneⅡA and deoxyschizandrin were 0.323 2-1.616 μg and 0.525-2.625 μg,respectively.The average recovery was 99.08% with RSD 0.42% for tanshinoneⅡA and 98.37% with RSD 0.73% for deoxyschizandrin respectively.Conclusion:The method is simple,accurate and rapid with good reproducibility.It can be used for the quality control of danshenwuweizi tablet.
