摘要
提出了转苏云金杆菌crylAb基因水稻抗螟性的快速离体叶片测定法,并用该方法评价了转crylAb基因水稻对二化螟、三化螟和大螟的抗性程度。结果表明,3种螟虫蚁螟取食纯合株系PR16及分离株系PR18、PR57或PR58中抗性植株叶片后,食叶量与对照和分离株系中的非抗性植株相比极显著下降,接虫后第4d的幼虫死亡率高达100%。该方法评价转crylAb基因水稻对3种螟虫的抗性结果与活体成株测定法评价结果完全吻合。离体叶片测定和活体成株测定均表明,纯合株系PR16对3种螟虫表现高抗性。
The 'detached-leaf bioassay', a rapid and easy method for determining the resistance of Bt (cry1Ab gene of Bacillus thuringiensis Berliner) transgenic rice to three stem borers, namely, striped stem borer Chilo suppressalis (Walker), yellow stem borer Scirpophaga incertulas (Walker) and purple stem borer Sesamia inferens (Walker), was developed and used to evaluate the resistance of different Bt transgenic rice lines to these stem borers. The results showed that after the newly hatched larvae of the three stem borers fed on the detached leaves of PR 16 (a homozygous Bt transgenic line) and resistant plants of PR18, PR57 and PR58 (the segregate Bt transgenic lines), their feeding amounts were significantly lower than those on the check variety (non-Bt-transgenic) and non-resistant plants of these segregate lines, all the former mortalities on the 4th day after inoculation were up to 100%. The test results of this method were quite coincident with those of the whole plant bioassay, both the two methods had indicated that the homozygous Bt transgenic line PR16 is highly resistant to the three stem borers.
