摘要
通过固体平板法,从食堂下水道污泥中筛选到1株耐热脂肪酶产生菌LJM-1,采用摇床培养法对该菌株的产酶条件进行了研究,结果表明,该菌株产脂肪酶的适宜培养基组成是:玉米粉淀粉6.0%﹑酵母粉3.5%﹑KH2PO40.3%﹑MgSO4.7H2O0.05%、(NH4)2SO40.5%﹑NaCl0.1%;适宜培养条件:培养温度40℃,初始pH7.7,摇床转速150r/min,装液量100mL/250mL三角瓶,接种量2.5%(体积分数),种龄72h,发酵时间48h。在此条件下,脂肪酶最高酶活为112.6U/mL。在单因素试验的基础上,采用正交试验对棉粕发酵脱毒条件进行了优化,得到最佳发酵条件是:初始pH7.7,培养温度40℃,接种量1.5%,发酵周期2d,酶活力达131.6U/mL。
A thermostable lipase producing strain LJM-1 was screened from canteen sewage sludge by solid plate method.The fementation conditions for lipase production were studied by using the methods of percussion table. The re- suits showed that the optimized culture conditions were as follows:corn starch 6.0%, yeast powder 3.5%, KH2PO 40.3%, MgSO4·7H2O 0.05%, (NH4)2SO4 0.5%, NaC1 0.1%, cultural temperature 40℃, the initial pH value 7.7, rotatory speed 150 r/min, 100 mL liquid medium in 250 mL erlemneyer flask, the vaccination quantity 2.5%(V/V), seed age 72 h, fer- mentation time 48 h. Under these conditions,the lipase avtivity reached 112.6 U/mL. Based on the single-factor test, he optimized fermentation conditions were investigated by orthogonal experimental design: the initial pH value 7.7, cultural temperature 40℃, the vaccination quantity 1.5%, fermentation time 2d, the lipase avtivity reached 131.6U/mL.
